5种外生菌根真菌培养特性研究

摘 要: 本研究测试了不同温度、pH、C源和N源对粘盖牛肝菌、绒粘盖牛肝菌、云绒盖牛肝菌、乳黄粘盖牛肝菌和褐环粘盖牛肝菌5种外生菌根真菌生长的影响。结果表明:1) 5种外生菌根真菌在5℃均不生长,乳黄粘盖牛肝菌、褐环粘盖牛肝菌生长的最适温度为30℃,粘盖牛肝菌为30℃~35℃,绒粘盖牛肝菌为20℃~25℃,云绒盖牛肝菌为20℃,褐环粘盖牛肝菌和粘盖牛肝菌在40℃也可生长,属于耐高温菌;2) 在测试pH3.0~pH9.0范围内,5种外生菌根真菌都可以生长,粘盖牛肝菌、云绒盖牛肝菌和褐环粘盖牛肝菌生长最适pH7.0,乳黄粘盖牛肝菌最适pH5.0,绒粘盖牛肝菌适宜pH5.0~pH7.0;3) 粘盖牛肝菌、云绒盖牛肝菌和褐环粘盖牛肝菌生长的最适C源为葡萄糖,乳黄粘盖牛肝菌对葡萄糖和麦芽糖利用程度相当,绒粘盖牛肝菌在5种C源上生长差异不显著;4) 粘盖牛肝菌、乳黄粘盖牛肝菌和褐环粘盖牛肝菌生长的最适N源为麦芽浸膏,绒粘盖牛肝菌为蛋白胨和麦芽浸膏,云绒盖牛肝菌对(NH4)2HPO4、蛋白胨和麦芽浸膏的利用程度相当。

Title: Study on culturing features of 5 ectomycorrhizal fungi

Abstract: The effects of different temperature, pH, carbon sources and nitrogen sources on the growth of five ectomycorrhizal fungi [Suillus bovinus (L. ex Fr.) Ktze., Suillus tomentosus (Kauff.) Sing. Snell & Dick, Xerocomus versicolor (Rostk.) Quél., Suillus lactifluus (With ex Gray) Sm. et Th. and Suillus luteus (L.ex Fr.)Gray] were tested. The results indicated that: 1) Five ectomycorrhizal fungi could not grow at 5℃, and the optimum growing temperature was 30℃, 30℃~35℃, 20℃~25℃, 20℃ for S. luteus and S. lactifluus, S. bovinus, S. tomentosus and X. versicolor, S. bovinus and S. luteus adapted to higher-temperature, because they could grow at 40℃; 2) All the ectomycorrhizal fungi were able to grow 1within the range of pH3.0~pH9.0, with the optimum pH value being pH7.0 for S. bovinus, X. versicolor and S. luteus, pH5.0 for S. lactifluus, and S. tomentosus had the same growth between pH5.0~pH7.0; 3) The optimum growing carbon sources for S. bovines, S. luteus and X. versicolor was glucose, as well as glucose and maltose had the same influence on the growth of S. lactifluus. The growth of S. tomentosus was not significantly different(P>0.05) on the 5 carbon sources; 4) The optimum growing nitrogen sources for S. bovines, S. lactifluus and S. luteus was malt extraction, together with peptone and malt extraction was the best for S. tomentosus, and ammonium dihydrogen phosphate, peptone and malt extraction had the same influence on the growth of X. versicolor.
