生脉胶囊对慢性心力衰竭大鼠心脏基因表达谱的影响

摘 要: 目的:探讨生脉胶囊对慢性心力衰竭大鼠心脏基因表达谱的影响。方法:将SD大鼠随机分为假手术组、心衰模型组和生脉胶囊组,采用腹主动脉缩窄7周复制大鼠慢性心力衰竭模型,分组灌胃治疗7周时,取大鼠心脏心尖部,运用Affymetrix大鼠全基因组表达谱芯片进行生脉胶囊组与心衰模型组心脏基因表达谱的差异性表达分析。结果:与心衰模型组比较,生脉胶囊组有333个基因表达上调,421个基因表达下调;其中差异表达基因共有115个(上调43个,下调72个),已知生物学过程的基因有51个(上调27个,下调24个),主要与代谢、信号转导、细胞周期及生长与增殖、心肌收缩、细胞粘附、免疫反应、转录、离子迁移、蛋白质转运等基因有关。结论:生脉胶囊可调整慢性心衰大鼠心脏基因表达谱,通过多途径、多环节、多靶点的方式发挥治疗作用。

Title: The effect of Shengmai capsule on the myocardium gene expressive profile in chronic congestive heart failure rats

Abstract: Objective: To study the effect of Shengmai capsule on the myocardium gene expressive profile in chronic congestive heart failure rats. Methods: SD female rats were divided randomly into 3 groups, 15 rats each group: sham-operation group (A), CHF model group (B), CHF model treated by Shengmai capsule group (C), preparing CHF rat models by constricting the suprarenal abdominal artery for 7 weeks. After 7 weeks treating respectively, took the apex of heart of the rats and analyzed the variability expression of the myocardium gene expressive profile between group C and group B with Affymetrix Rat Expression Array 230 2.0 Gene Chip. Results: Compared with the CHF model group (B), 333 gene expressions were up-regulated in Shengmai capsule group (C), other 421 down-regulated. There were totally 115 gene variability expression (43 up-regulated and 72 down-regulated). Thereinto, 51 genes (27 up-regulated and 24 down-regulated), which biology process were clear, were related to metabolism, signal transduction, cell cycle, cell growth and progression, striated muscle contraction, cell adhesion, immune response, transcription, ion transport, protein transport and so on. Conclusion: Shengmai capsule could regulate the gene expression profile of CHF rats’ myocardium, and produce a marked therapeutic effect through ways of multi-avenue, multi-link, and multi-target.
