Pokemon对p14ARF→Mdm2→p53途径的影响

摘 要: p14ARF→Mdm2→p53致癌途径广泛存在于人类各种肿瘤形成过程,p14ARF和p53是最常见的抑癌基因,p53是细胞周期调节因子,活化的p53蛋白可以促进编码p21蛋白靶基因的转录,使细胞滞留于G1期;p53还可促发DNA损伤的细胞凋亡;Mdm2可以限制p53的反应程度,并可直接抑制p53转录,还可调节p14ARF的功能;p14ARF负性调节Mdm2,从而间接调节p53转录。而p53反过来又可以影响Mdm2和p14ARF的表达。本综述就这条反馈环路的各个部分及其相互关系进行了详细的阐述,并着重就Pokemon基因对p14ARF→Mdm2→p53的影响做了分析,最新的研究显示Pokemon基因可以作为对癌症进行基因治疗的新的有效靶基因。

Abstract: p14ARF→Mdm2→p53 pathway is commonly existed in human tumor progression. p14ARF and p53 are most common tumor suppressor genes, both of them are cell cycle modulators. The activated p53 protein could promote the transcription of target gene, by which the p21protein was encoded, thus inhibit the cell at G1 stage; p53 also could induce the cell with damaged DNA into apoptosis; Mdm2 could limit the duration and activity of p53 in response to stress, and inhibit the transcription of p53 directly, regulate the function of p14ARF; P14ARF negatively regulate the Mdm2, thus adjust the transcription of p53 indirectly. Conversely p53 affect the expression of Mdm2 and p14ARF. In this review article, all of the components in the p14ARF→Mdm2→p53 pathway and their complicated interaction are discussed, and the effect of the pathway by Pokemon is emphasized. The latest research suggest that Pokemon may offer an effective new target for cancer therapeutics.
