猪miR-29c的克隆、组织表达谱及与部分免疫性状的关联分析

摘 要: 研究利用stem-loop RT-PCR法克隆猪miR-29c成熟序列,通过实时定量PCR技术检测出猪miR-29c在心、肝、脾、肺、肾、肌肉、胃、小肠等8种组织中广泛表达,其中在心脏、脾脏、肾脏中高表达。测序发现在miR-29c前体序列上游32 nt处有一个T/C突变,利用PCR-限制性片段长度多态性(restricted fragment length polymorphisms, RFLP)技术进行多态性检测及部分血液参数和免疫性状关联分析。结果显示:NcoⅠ酶切基因型与长白猪群体的出生重、0日龄(出生)的平均红细胞体积(mean red cell volume,MCV)、17日龄的血小板计数分布宽度(platelet distribution width,PLDW)和32日龄猪瘟抗体阻断率存在显著相关(P<0.05);与0日龄的平均血红蛋白量(mean corpuscular hemoglobin,MCH)、0日龄和32日龄的平均血红蛋白浓度(mean corpuscular hemoglobin concentration, MCHC)存在极显著相关(P<0.01)。位于前体的单核苷酸多态性(single nucleotide polymorphism,SNP)可能调节Drosha的加工过程,从而影响miRNA的表达水平。

Title: Association analysis on cloning, organization expression pattern and part immune traits of porcine miR-29c

Abstract: In this study, the mature sequence of porcine miR-29c was cloned by stem-loop RT-PCR. It is widely expressed in 8 pig tissues, including heart, liver, spleen, lung, kidney, muscle, stomach, and small intestine by real-time PCR assays. One T/C polymorphism at the position 32 nt of miR-29c precursors was tested in a Landrace population by PCR-RFLP and analyzed association with immune traits. The results show that NcoⅠ enzyme genotype is significantly associated (P<0.05) with birth weight, the mean red cell volume (MCV) of 0-day-old pigs; the platelet distribution width (PLDW) of 17-day-old pigs and block of classical swine fever antibodies of 32-day-old pigs, and is more significantly associated (P<0.01) with the mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) of 0-day-old pigs, the MCHC of 32-day-old pigs. The single nucleotide polymorphism in precursor can modulate Drosha processing efficiency and thereby affect the level of miRNA expression.
