基于环糊精采用主客体化学法制备的基因载体

摘 要: 综述了应用主客体化学方法、以环糊精为主体制备用于基因治疗的非病毒基因载体。不同的客体分子与环糊精通过非共价作用结合成不同形状略带正电荷的复合物,通过静电作用吸附带负电荷的DNA或siRNA,形成阳离子纳米粒经胞吞进入靶细胞,快速从溶酶体中逃逸并释放目的基因到靶向位点,从而改善基因载体的性能和提高药效。讨论了不同外界条件对环糊精基因载体在细胞内逃逸与释放的影响,并展望了未来以环糊精为主体的主客体化学用于构建基因载体的广阔前景。

Title: Gene vector based on cyclodextrin by using host guest inclusion chemistry

Abstract: In this paper, we reviewed non-viral gene vector based on cyclodextrin by using host-guest inclusion chemistry. The delivery efficiency and performance of gene vector can be improved through non covalence inclusion between different guest molecules and cyclodextrin, which combine into slightly positive charge complexes of different shapes, interacting with the negatively charged DNA or siRNA through electrostatic interaction. Cationic nanoparticles enter the target cells, then escape from the endosomes and release the target gene into nuclear. The effect of different outside condition on intracellular release of gene vector based on cyclodextrin was discussed. The wide perspective that gene vectors formed by using host-guest inclusion chemistry based on cyclodextrin was prospected.
