辣椒碱预防大鼠营养性肥胖作用的研究

摘 要: 目的:观察辣椒碱(capsaicin,CAP)对超重大鼠体重、体脂含量及血糖血脂的影响,探讨CAP预防营养性肥胖的作用。方法:将40只大鼠随机分为正常对照组(ND)、模型组(HD)和CAP低(HD+5)、中(HD+10)、高剂量(HD+20)干预组,利用高糖高脂饲料诱导建立营养性超重大鼠模型,CAP干预4周,观察其体重、内脏脂肪湿重的变化。采用酶法检测血清葡萄糖(glucose,GLU)、甘油三酯(triglyceride,TG)和总胆固醇(total cholesterol,TC),选择性沉淀法检测低密度脂蛋白胆固醇(low density lipoprutein-cholesterol,LDL-C)和高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)。结果:CAP各剂量组大鼠体重低于模型组;中剂量组内脏脂肪湿重低于模型组,高剂量组内脏脂肪湿重、内脏脂肪/体重均低于模型组(P<0.01);与模型组相比,各剂量组大鼠血清TC,LDL-C,TG及GLU水平降低,而HDL-C水平升高(P<0.01),并呈剂量-效应关系,但高剂量组大鼠各项指标与正常对照组相比无显著差异。结论:CAP可有效抑制超重大鼠体重的增加,减少体脂聚集,调节血糖血脂水平,对营养性肥胖有显著的预防作用。

Title: Prophylactic effect of capsaicin on alimentary obesity of rats

Abstract: Objective: To observe the effects of capsaicin (CAP) on body fat, plasma lipids and glucose in overweight rats and investigate the prophylactic effect of CAP on alimentary obesity. Methods: 40 rats were divided randomly into five groups: normal-control group (ND), model group (HD), low CAP supplement group (HD+5), middle CAP supplement group (HD+10) and high CAP supplement group (HD+20). The rats were fed with high fat and carbohydrate diet to induce overweight, and then treated with CAP for four weeks. We observed the changes of body weight and wet weight of the rat visceral fat. The plasma glucose (GLU), triglycerides (TG) and total cholesterol (TC) were assayed with enzymic method. Low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) were assayed with selective precipitation method. Results: The body weight of CAP treated groups was lower than that of the HD group. The wet weight of visceral fat of HD+10 group was lower than that of the HD group, and the wet weight of visceral fat and the visceral fat/body weight of HD+20 were lower than that of the HD group (P<0.01). Compared with the HD group, the plasma TC, LDL-C, TG and GLU were reduced in CAP treated groups, while the HDL-C in these three groups were increased (P<0.01), and there was a dose dependent relationship in the CAP treated groups. However, there was no significant difference of each index between HD+20 group and ND group. Conclusion: CAP may reduce the body weight, inhibit the concentration of body fat, and regulate the level of plasma lipids and glucose in overweight rats. Therefore, CAP has a beneficial effect on the prophylaxis of the alimentary obesity.
