碘缺乏对大鼠仔鼠海马凋亡的影响

摘 要: | 目的:观察碘缺乏对大鼠仔鼠海马凋亡的影响。方法:健康2月龄雌性Wistar大鼠(n=14),交配妊娠后,取孕鼠7只,按体重随机分成对照组和碘缺乏组。自妊娠第6天起至生后第28天(postnatal day 28,PND28),碘缺乏组饲以缺碘饲料,饮用自来水,对照组饲以普通饲料。分别于PND14,PND21和PND28时,每组随机取5只仔鼠,灌流固定大脑,进行TUNEL染色,观察海马凋亡细胞。结果:PND14和PND21时,碘缺乏组仔鼠海马各区TUNEL阳性细胞显著高于对照组(P<0.05)。在PND28时,各组均未发现TUNEL阳性细胞。结论:碘缺乏可造成仔鼠海马凋亡细胞增加,凋亡峰延迟。 |

Title: | Effects of iodine-deficient on apoptosis in hippocampus of pups |

Abstract: | Objective: To observe the effects of iodine deficiency on apoptosis in hippocampus of pups. Methods: Two-month old heathy female Wistar rats (n=14) after pregnancy were randomly divided into control group and iodine-deficient group (7 in each group). From gestational day 6 to postnatal day (PND) 28, iodine-deficient group was administered with iodine-deficient diet and tap water, while control group was fed with normal diet. 5 pups from each group were sacrificed and intracardiac perfused in PND14, PND21 and PND28 respectively. Brains were removed,fixed and sectioned coronally. The apoptosis in the hippocampus was determined by TUNEL assay. Results: In PND14 and PND21, the apoptosis cells was increased in the iodine-deficient group in hippocampus (P<0.05). In PND28, the positive cells were scarcely observed in all groups. Conclusion: Iodine deficiency may result in the increase of apoptosis cells and delay of apoptosis peak in hippocampus of pups. |
