新加良附方联合5-Fu对裸鼠胃癌移植瘤survivin/caspase-3表达的影响

摘 要: 目的:观察新加良附(Xinjialiangfu,XJLF)方联合5-氟尿嘧啶(5-Fu)对人胃癌细胞(SGC-7901)裸鼠移植瘤生长抑制作用及其对肿瘤组织中survivin及caspase-3蛋白表达的影响。方法:建立移植性人胃癌细胞(SGC-7901)裸鼠移植瘤模型,随机分为模型对照组,5-Fu组及XJLF方高、中、低剂量组,联合给药组(XJLF中剂量+5-Fu组)等6组。连续给药10 d,计算肿瘤抑制率,检测survivin/caspase-3蛋白的表达。结果:联合给药组抑瘤率为60.28%,与模型对照组比较,有统计学意义(P<0.05);与5-Fu组和XJLF高剂量组比较,联合给药组抑瘤率升高明显,有统计学意义(P<0.05);联合给药组上调肿瘤组织caspase-3蛋白表达,下调survivin蛋白表达,与模型对照组比较,有统计学意义(P<0.05,P<0.01);与5-Fu组和XJLF高剂量组比较,联合给药组caspase-3蛋白表达明显升高(P<0.05),survivin蛋白表达明显下降(P<0.05)。结论:联合给药组抑瘤率高于XJLF高剂量组及5-Fu组,在调节caspase-3,survivin表达方面优于XJLF高剂量组及5-Fu组,显示了两药协同作用的优势。

Title: Effects of Xinjialiangfu formula and 5-Fu combination on survivin and caspase-3 expression of gastric cancer xenografts in nude mice

Abstract: Objective: To observe the effect of Xinjialiangfu (XJLF) formula on human gastric cancer (SGC-7901) tumor growth in nude mice and its impact on survivin and caspase-3 expression in tumor tissue. Methods:The animal model of SGC-7901 xenografted nude mice was established, and all the mice were randomly divided into 6 groups namely model control group, 5-Fu group, XJLF formula high-dose group, XJLF formula medium-dose group, XJLF formula low-dose group and drug combination group (XJLF formula medium-dose and 5-Fu). After 10 d of continuous treatment, tumor inhibition rate was calculated and the expression of survivin and caspase-3 was detected by immunohistochemistry techniques. Results: The tumor inhibition rate of drug combination group was 60.28%, which was significantly increased compared to the model control group (P<0.05), 5-Fu group (P<0.05) and XJLF high-dose group (P<0.05), respectively. Meanwhile, the caspase-3 expression was significantly up-regulated and the survivin expression was significantly down-regulated in drug combination group, compared to the model control group (P<0.05,P<0.01), 5-Fu group (P<0.05,P<0.05) and XJLF formula high-dose group (P<0.05,P<0.05), respectively. Conclusion: The tumor inhibition rate of drug combination group is higher than XJLF formula high-dose group and 5-Fu group. Also combination group has stronger effect compared to model control group in terms of own-regulation of caspase-3 expression and up-regulation of survivin expression, which indicates a benefit of XJLF and 5-Fu combination treatment.
