高糖对心房肌钙信号的影响及机制

摘 要: 目的:探讨高糖对大鼠心房肌钙信号的影响及机制。方法:分离雄性SD大鼠心房肌细胞并行原代培养。分成两部分:第一部分分为4组,1)左旋葡萄糖组(LG组);2)正常葡萄糖组(NG组);3)高葡萄糖组(HG组);4)HG+尿酸(HG+U)组。第二部分分为3组,1)ONOO�组;2)DC-ONOO�组;3)ONOO� + U组。观察钙信号、钙通道RYR2 mRNA及其上游调控因子FKBP12.6 mRNA的表达。结果:HG组的钙瞬变幅度高于NG组,ONOO�可促进钙的释放。尿酸可减弱ONOO�对钙释放的促进作用。HG 组RYR2 mRNA的表达较NG组明显下降。HG+U组RYR2 mRNA的表达较HG组明显上升。ONOO�组降低RYR2 mRNA的表达。HG 组RYR2调控因子FKBP12.6 mRNA的表达明显下降。结论:高糖可促进心房肌细胞钙的释放,其机制可能与高糖氧化应激影响钙通道有关。

Title: Effects and mechanism of high glucose on calcium signaling in atrial myocytes

Abstract: Objective: To investigate the effect of high glucose on calcium signals of atrial myocytes and its mechanism. Methods: The experiment based on isolated primary cultured SD atrial myocytes rats were divided into two parts, the first section was divided into four groups: 1) L-glucose (LG) group; 2) normal group (NG); 3) high glucose (HG) group; 4) HG+uric acid (HG+U) group. The second section was divided into three groups: 1) ONOO� group; 2) DC-ONOO� group; 3) ONOO�+U group. We observed the change of calcium signaling, calcium channel RYR2 mRNA and the expression of its upstream regulatory gene FKBP12.6 mRNA. Results: Calcium transient amplitude was larger in the HG group compared with the NG group. Application of uric acid significantly decreased the calcium transient amplitude whereas exposure of myocytes to ONOO� markedly increased. The expression of RYR2 mRNA in the HG group was decreased when compared to the NG group. Incubation in uric acid increased the expression of RYR2 mRNA, whereas ONOO� decreased the amount of RYR2 mRNA. The expression of FKBP12.6 mRNA was decreased in the HG group compared to NG treated cells. Conclusion: High glucose increases the release of calcium, the mechanism may be associated with high glucose-induced oxidative stress on calcium channel.
