S180荷瘤小鼠肝脏细胞的DANCE反应及其机制

摘 要: 目的:在S180荷瘤病理变化的条件下,研究小鼠肝脏细胞发生的非生理性细胞内密度改变(density alteration in non-physiological cells,DANCE)反应,探讨小鼠肝脏细胞DANCE反应的发生机制。方法:构建S180荷瘤小鼠模型,以不连续蔗糖密度梯度离心法(discontinuous sucrose gradient centrifugation,DSGC)测定种瘤后不同时间小鼠肝脏细胞的DANCE反应,并以ELISA、全自动生化分析仪检测不同时间S180荷瘤小鼠的血清C-反应蛋白(C-reactive protein,CRP)、谷丙转氨酶( alanine aminotransferase,ALT)水平变化。结果:接种S180肿瘤细胞后,在不同时间小鼠肝脏细胞发生不同程度的DANCE反应,其中,在接种肿瘤细胞后的第10 天和第25天,小鼠肝脏细胞的DANCE反应概率最高,分别为4/7和7/7. ELISA和全自动生化分析仪检测结果显示,S180荷瘤小鼠血清CRP,ALT水平均较对照组的高,且具有显著性差异。结论:在S180肿瘤形成与发展的病理过程中,小鼠肝脏细胞发生显著的DANCE反应,而且血清CRP,ALT水平亦升高,提示至少部分肝脏细胞发生免疫或炎症反应,这可能是肝脏细胞发生DANCE反应的机制之一。

Title: DANCE in tumor-bearing mouse liver cells and the mechanism

Abstract: Objective: To study density alteration in non-physiological cells (DANCE) in mouse liver cells caused by pathological changes after transplantation of S180 tumor in mice, and thereby to explore the mechanisms of DANCE in mouse liver cells in vivo. Methods: A mouse model of S180-transplanted tumor was constructed, and DANCE in the liver cells was determined by discontinuous sucrose gradient centrifugation (DSGC) on every 5 days after the tumor transplantation. ELISA, automatic biochemical analyzer were employed to determine the serum levels of C-reactive protein (CRP) and alanine aminotransferase (ALT) in the tumor-baring mice. Results: S180 tumor-bearing mice show that DANCE occurrs in the liver cells with different occurrence rates at different time points after the tumor transplantation. The incidence of DANCE in the mouse liver cells reaches two peaks, as high as 4/7 and 7/7, at the time of tenth and twenty-fifth day after the tumor-transplantation, respectively. Meanwhile, the serum levels of CRP and ALT in tumor-bearing mice are found to be significantly higher than those in the control group. Conclusion: Transplantation of S180 tumor in mice may result in DANCE in the liver cells, and increase serum levels of CRP and ALT in the tumor-bearing mice may indicate immune response or inflammation in, at less part of the mouse liver cells, which may be one of the mechanisms for occurrence of DANCE in the tumor-bearing mouse liver cells.
