阿霉素涂层输尿管支架的制备及体外药物释放行为研究

摘 要: 采用开环共聚法,以异辛酸亚锡为催化剂制备聚乳酸−聚己内酯[poly(L-lactide-co-ε-caprolactone),PLCL5050]共聚物。通过核磁共振氢谱(1H nuclear magnetic resonance,1H NMR)、核磁共振碳谱(13C NMR)和凝胶渗透色谱(gel permeation chromatograph,GPC)表征共聚物的结构,通过差式扫描量热法(differential scanning calorimetry,DSC)、万能力学试验机表征共聚物的热力学和力学性能。采用溶液浸渍法制备阿霉素(doxorubicin,DOX)涂层输尿管支架,并通过环氧乙烷(ethylene oxide,EtO)灭菌方式进行灭菌。研究DOX涂层支架在人工尿液中的药物释放行为,探讨载药量和EtO灭菌处理对DOX药物释放行为的影响。结果表明,载药量的增加会降低药物的释放速度,而EtO灭菌会使DOX释放速度加快。

Title: Preparation and in vitrodrug release study of doxorubicin-coated ureteral stents

Abstract: Poly(L-lactide-co-ε-caprolactone) (PLCL5050) copolymer was synthesized successfully via ring-opening polymerization by using stannous iso-caprylate as the catalyst. 1H nuclear magnetic resonance (1H NMR), 13C NMR and gel permeation chromatograph (GPC) were applied to characterize the structure of the copolymer. The thermal and mechanical properties of PLCL5050 were measured by differential scanning calorimetry (DSC) and tensile tests. PLCL5050 as carriers for doxorubicin (DOX) was coated on ureteral stents by the solution dipping method. Both non-sterilized and ethylene oxide (EtO) sterilized stents were used for loading DOX. To simulate a body environment, the release profiles of DOX-coated ureteral stents were evaluated in artificial urine, and the effects of the drug does and sterilization were investigated. The results indicatedthat the release rate of DOX decreased with the increasing drug loading, while increased when the stents were sterilized by EtO.
