thalidomide通过保护软骨下骨延缓小鼠DMM骨关节炎

摘 要: 目的:观察thalidomide在小鼠内侧半月板不稳定(destabilization of the medial meniscus,DMM)骨关节炎模型中对软骨和软骨下骨的影响,为阐述其在骨关节炎发展进程中的作用提供依据。方法:将C57/BL6小鼠随机分为实验组、对照组和假手术组,实验组、对照组小鼠右膝关节均采用DMM方法建立膝关节骨关节炎模型,假手术组则只在右膝关节相应位置切开皮肤和关节囊。从术后第1天起,实验组用thalidomide腹腔注射给药,对照组和假手术组按相同标准注射生理盐水。采用基质金属蛋白酶-13(matrix metalloproteinase-13,MMP-13)免疫荧光、番红O-固绿染色、微计算机断层扫描技术(micro-computed tomography,micro-CT)和苏木精-伊红(hematoxylin-eosin,HE)染色分别检测各组小鼠膝关节软骨和软骨下骨变化。结果:2周时,实验组和假手术组软骨下骨变化明显小于对照组,各组小鼠关节软骨变化无明显差异;4周时,实验组软骨下骨和关节软骨变化均明显小于对照组,假手术组软骨下骨和关节软骨无明显变化。结论:thalidomide能减少DMM骨关节炎模型中软骨下骨的病理性改变,可能对延缓骨关节炎的发展起着重要作用。

Title: Thalidomide inhibition of DMM osteoarthritis in mice via protecting subchondral bone

Abstract: Objective: To investigate the effects of thalidomide on cartilage and subchondral bone in destabilization of the medical meniscus (DMM) osteoarthritis model in mice, and to provide evidence for its role in the development of osteoarthritis. Methods: The C57/BL6 mice were randomly divided into experimental group, control group and sham operation group. DMM method was used on the mice of the experimental group and the control group to establish the model of knee osteoarthritis, the sham operation group merely cut the skin and joint capsule in the corresponding position of the right knee joint. The experimental group was injected intraperitoneally with thalidomide, and the control group and sham operation group were injected with normal saline according to the same standard in the first day after the surgery. Matrix metalloproteinase-13 (MMP-13) immunofluorescence, safranin O-fast green staining, micro-computed tomography (micro-CT) and hematoxylin-eosin (HE) staining were used to detect the changes in cartilage and subchondral bone. Results: At 2 weeks, the changes in subchondral bone in the experimental group and sham operation group were significantly less than the control group, there was no significant difference in articular cartilage of mice in each group. At 4 weeks, the changes in subchondral bone and articular cartilage in the experimental group were significantly less than those in the control group, there was no obvious change in subchondral bone and articular cartilage in the sham operation group. Conclusion: Thalidomide can reduce the pathological changes of subchondral bone in DMM osteoarthritis model, and may play an important role in delaying the development of osteoarthritis.
