蒙古白丽蘑子实体下土壤菌根真菌与 深色有隔内生真菌

摘 要: 结合形态学鉴定与分子鉴定研究蒙古白丽蘑子实体下土壤菌根真菌的多样性,同时也从形态学角度对深色有隔内生真菌(dark septate endophytes,DSE)进行研究。结果如下:1)通过形态学鉴定,可得3 种初步鉴定到种的丛枝菌根(arbuscular mycorrhizal,AM)真菌——缩球囊霉(Glomus constrictum)、孔窝无梗囊霉(Acaulospora foveata)和格氏原囊霉(Archeospora gerdemannii);2)AM 真菌对根部侵染的结构有根外菌丝,侵入点,根外孢子,胞内菌丝,胞间菌丝,Paris 型丛枝,以及棒状、不规则型和成串型3 种类型的泡囊;3)DSE 的结构有菌丝横隔、微菌核和孢子,且分布于根细胞表面、细胞间和细胞内;4)对3 个土壤样品进行高通量测序,得到6 个在各样品中分布差异明显的外生菌根(ectotrophic mycorrhiza,ECM)真菌可操作分类单元(operational taxonomic unit,OTU),结合系统发育分析,其中2 个只鉴定到柔膜菌目(Helotiales),其余4 个分别鉴定到了棉革菌属(Tomentella)、红菇属(Russula)、空团菌属(Cenococcum)和Amphinema. 本文首次研究了蒙古白丽蘑子实体下土壤菌根真菌及DSE,这可能有助于从共生关系的角度摸索蒙古白丽蘑生长的相关条件,并为蒙古白丽蘑的驯化栽培提供研究基础。

Title: Mycorrhizal fungi and dark septate endophytes in the soils under the fruit bodies of Leucocalocybe mongolicum

Abstract: Morphological and molecular identification had been combined to investigate the diversity of mycorrhizal fungi in the soils under fruit bodies of Leucocalocybe mongolicum, and dark septate endophytes (DSE) had also been studied morphologically at the same time. The results are: 1) Three species of arbuscular mycorrhizal (AM) fungi have been identified morphologically, Glomus constrictum, Acaulospora foveata, and Archeospora gerdemannii; 2) The structures of AM fungi to infect the roots are hyphae on root surface, external hyphae, entry point, external spore, intracellular hyphae, intercellular hyphae, Paris-type arbuscule and three types of vesicles (clavate, irregular and string); 3) Hyphae, microsclerotia and spores of DSE distributed on the surface, intercellular and intracellular of root cells have been observed; 4) Six ectotrophic mycorrhiza (ECM) operational taxonomic units (OTUs) in each sample had been identified by the high-throughput sequencing, and the distributions of them in samples are obvious, through phylogenetic analysis, two of them belong to Helotiales, the others belong to genera of Tomentella, Russula, Cenococcum and Amphinema. This is the first report on the mycorrhizal fungi and DSE in the soils under the fruit bodies of L. mongolicum, and it can explore the relevant conditions of L. mongolicum growth on the angle of symbiotic relationship and provide basical research on domestication and cultivation of L. mongolicum.
