水凝胶表面微结构对排异反应的调控

摘 要: 为研究水凝胶表面微结构对排异反应的影响与调控,基于改进的“微球模板法”,通过对聚甲基丙烯酸甲酯(poly(methyl methacrylate),PMMA)微球模板进行处理,在多孔水凝胶表面构建了宽度为250 μm、类似“条形码”的实心条带,成功调控了表面实心条带的间距,并利用小鼠皮下移植实验考察不同表面微结构引起的体内急性和慢性排异反应。动物实验表明,当水凝胶表面相邻实心条带的间距不同时,对应的巨噬细胞聚集状况和胶原包封膜的形成情况均明显不同,说明通过水凝胶表面微结构的调节成功实现了对排异反应的调控。本研究为进一步深入探索表面微结构与排异反应的相关性以及免疫行为的调控提供了新思路和新方法。

Title: Modulation of the foreign body reaction by hydrogel surface microstructures

Abstract: To study the effect and modulation of the foreign body reaction (FBR) by hydrogel surface microstructures, the porous hydrogel materials with different “barcode-like” surface microstructures were developed by processing the poly(methyl methacrylate) (PMMA) sphere templates based on modified “sphere templated method”. The surfaces of constructed materials possessed nonporous strips with a width of 250 μm, and the distances among them were tunable. The surface microstructures-induced acute and chronic FBR were tested by mice subcutaneous implantation experiment. The results showed that the conditions of macrophage cell accumulation and collagenous capsule formation changed dramatically with different distances among nonporous strips of hydrogel surface, and demonstrated that the modulation of the FBR was successfully achieved by hydrogel surface microstructure control. This work provides new ideas and methods for the further exploration of correlation between surface microstructures and FBR as well as the regulation of immune activity.
