自组装诱导金纳米粒子聚集用于癌症细胞的光热治疗

摘 要: 为解决一些小分子药物水溶性差、无靶向等缺点,纳米载药体系应运而生,而又因为癌症病因的复杂性,单纯的化学药物治疗不能达到良好的治疗效果,所以联合治疗的方式越来越引起研究人员的兴趣。光热治疗(photothermal therapy,PTT)具有时间空间双重可控性,利用生物相容性良好的材料在光照后本身产生的热量可造成肿瘤细胞凋亡。研究利用萘乙酸衍生物(NA-DADO)和金纳米粒子(AuNPs)接β-环糊精(Au-CD)的主客体作用,使AuNPs在水溶液中发生聚集。之后研究聚集状态下金纳米粒子(Au-Aggr)的光热效应。发现用808 nm的近红外光(near-infrared ray,NIR)照射5 min时,Au-Aggr溶液温度最高能上升21℃. 体外细胞毒性结果显示,Au-Aggr对于肿瘤细胞具有良好的杀伤性,且由于体系中透明质酸(HA)的存在,对于表面含有透明质酸受体的B16F10细胞的治疗效果最好。因此,本方法制备得到的Au-Aggr在肿瘤治疗方面具有显著的疗效。

Title: Self-assembly induced gold nanoparticle aggregation for photothermal therapy of cancer cells

Abstract: In order to solve the shortcomings such as poor water solubility and non-targeting of some small-molecule drugs, the nanoparticle drug delivery system comes into being. And because of the complexity of the cancer etiology, simple chemical drug therapy cannot achieve a good therapeutic effect, more and more researchers are interested in combination therapy. Photothermal therapy (PTT) has spatial and temporal controllability, good biocompatible materials generate heat to cause apoptosis of tumor cells by irradiation. Because of the host-guest interaction between naphthaleneacetic acid derivatives (NA-DADO) and β-cyclodextrin gold nanoparticles (Au-CD), the aggregation of gold nanoparticles (AuNPs) was observed in aqueous solution. Afterwards, the photothermal effect of aggregation of gold nanoparticles (Au-Aggr) was studied. The temperature of the Au-Aggr solution rises 21℃ with near-infrared ray (NIR) irradiation at 808 nm for 5 min. The results of in vitro cytotoxicity showed that Au-Aggr had a good killing effect on tumor cells. And due to the presence of hyaluronic acid (HA) in the system, the treatment of B16F10 cells with HA receptor on the surface was the best. Therefore, the Au-Aggr prepared in this paper had a significant effect in the treatment of tumors.
