柠檬酸生产菌株改造研究进展

摘 要: 中国是世界上最大的柠檬酸(citric acid,CA)生产和出口国。提高CA 的发酵效率对加强我国CA 产业的国际竞争力具有重大意义,提高CA 的生产效率关键在于获得高效的CA 生产菌株。本研究总结了近年来各国学者在CA 生产菌株改造方面的研究成果,主要侧重于总结工业化生产中利用的两类主流菌株——黑曲霉(Aspergillus niger)和耶罗威亚属(Yarrowia sp.)酵母菌的改造进展,阐述了包括物理化学诱变和基因工程改造在内的主流改造策略。目前,物理化学诱变的育种方法主要用于A. niger 的改造,而对耶罗威亚酵母菌(Y. lipolytica)的改造主要利用基因工程的方法;随着A. niger 遗传操作系统研究的发展,近年来已有很多对A. niger 进行基因工程改造的报道。此外,本研究还进一步展望了在当前系统生物学迅猛发展的背景下,CA生产菌株研发的新趋势。

Title: Research progress of strain improvement for citric acid production

Abstract: The largest production and exportation of citric acid (CA) are in China. It is very important to enhance the fermentation efficiency of CA for a dominant position in the global competition. It is also the key point to screen highly efficient CA producing strain for improving the production efficiency of CA. This paper summarizes the recent research development for enhancing CA producing strains in recent years, including physical and chemical mutagenesis and genetic engineering advances. The main strategies for achieving high yields of CA are summarized in this paper. This manuscript mainly focuses on the improvement progress of two kinds of strains, including Aspergillus niger and yeast genus of Yarrowia sp., which are mainly used in industrial CA production. Now, physical and chemical mutagenesis methods are mainly used for improving A. niger, while the modification of Y. lipolytica mainly use genetic engineering. With the development of genetic operating system of A. niger, there have been many reports on genetic engineering for improving A. niger in recent years. Furthermore, we also prospect the new trends of researches on the further development of CA producing strains promoted by the current rapid development in systems biology.
