新颖栽培丹参材料的rDNA内外转录间隔区(ITS和ETS)分子标记鉴定

摘 要: 本研究对前期发现的2个新颖多年生大紫花单株(V-HNYZ-bV-1和W-SXXA-bV-1)和1个二年生白花丹参(Salvia miltiorrhiza)栽培居群(W-SCHY-W-1)材料,进行了基于其rDNA内、外转录间隔区(ITS和ETS)的分子标记鉴定。结果表明:W-SXXA-bV-1和V-HNYZ-bV-1的ITS的GC含量均较高,W-SCHY-W-1的ITS和ETS的GC含量最低;新颖材料W-SXXA-bV-1和V-HNYZ-bV-1的ITS序列变异率较高,均高于W-SCHY-W-1,而对于ETS序列,W-SXXA-bV-1和V-HNYZ-bV-1接近(7.1%~6.5%),但低于W-SCHY-W-1(8.2%);W-SXXA-bV-1与V-HNYZ-bV-1亲缘关系较近,W-SXXA-bV-1、V-HNYZ-bV-1和W-SCHY-W-1均与Q-DZH-V-4亲缘关系较远,V-HNYZ-V-1与Q-DZH-V-4亲缘关系很近。上述结果与栽培形态观察结果都高度一致表明:W-SXXA-V-1和V-HNYZ-V-1是康定或甘西鼠尾草,新颖单株材料W-SXXA-bV-1可能为W-SXXA-V-1的突变体,V-HNYZ-V-1和Q-DZH-V-4同为荔枝草,V-HNYZ-bV-1可能为混杂在V-HNYZ-V-1中的康定或西藏鼠尾草,与W-SXXA-bV-1非常相似。W-SCHY-W-1为粘毛鼠尾草。

Title: Molecular marker identification of rDNA internal and external transcribed spacer (ITS and ETS) of novel cultivated Danshen materials

Abstract: In this research, two novel perennial large purple-flowered monocots (V-HNYZ-bV-1 and W-SXXA-bV-1) and one biennial white-flowered Danshen (Salvia miltiorrhiza) in field cultivation (W-SCHY-W-1) were identified by molecular marker based on the internal and external transcribed spacers (ITS and ETS) of the rDNA. Results showed that the GC contents of ITS in W-SXXA-bV-1 and V-HNYZ-bV-1 were relatively high, while the GC contents of ITS and ETS in W-SCHY-W-1were the lowest. The ITS sequence variation rates of the novel materials W-SXXA-bV-1 and V-HNYZ-bV-1 was relatively high, and significantly higher than that of W-SCHY-W-1. The ETS sequence variation rates of W-SXXA-bV-1 and V-HNYZ-bV-1 were closer (7.1%-6.5%), but lower than that of the W-SCHY-W-1 (8.2%). The relationship between W-SXXA-bV-1 and V-HNYZ-bV-1 was closer, the relationship between W-SXXA-bV-1/V-HNYZ-bV-1/W-SCHY-W-1 and Q-DZH-V-4 was relative distant, and the relationship between V-HNYZ-V-1 and Q-DZH-V-4 was the closest. The results were highly consistent with field cultivational observation. It is suggested that W-SXXA-V-1 and V-HNYZ-V-1 are S. prattii or S. przewalskii. The novel single plant W-SXXA-bV-1 is likely a mutant of W-SXXA-V-1. Both V-HNYZ-V-1 and Q-DZH-V-4 are S. plebeian. V-HNYZ-bV-1 is probably a material of S. prattii or S. wardii promiscuous in V-HNYZ-V-1, which is very similar to W-SXXA-bV-1. W-SCHY-W-1 is S. roborowskii.
