黄芩总黄酮纯化工艺研究

摘 要: 目的:通过纯化工艺研究,优选比较黄芩总黄酮的纯化方法。方法:以紫外分光光度法及高效液相色谱法为分析手段,以黄芩总黄酮、黄芩苷、汉黄芩素含量及转移率为考察指标,对酸沉法与HP20大孔吸附树脂纯化2种方法进行对比研究。结果:以酸沉法纯化黄芩总黄酮,黄芩总黄酮的纯度为97.61%,黄芩苷、汉黄芩素含量分别为9.19 g/100 g生药,0.71 g/100 g生药,三者转移率分别为99.33%,90.05%,82.96%;以HP20大孔吸附树脂纯化黄芩总黄酮,黄芩总黄酮纯度为80.66%,黄芩苷、汉黄芩素含量分别为8.97 g/100 g生药,0.53 g/100 g生药,三者转移率分别为84.01%,78.21%,77.24%.结论:酸沉法纯化黄芩总黄酮,其黄芩总黄酮、黄芩苷、汉黄芩素的含量及转移率明显高于HP20大孔吸附树脂法,黄芩总黄酮纯化方法以酸沉法为好。

Title: Study on purification processes of total flavone in Scutellaria baicalensis Georgi

Abstract: Objective: To select a preferable purification method of total flavone in Scutellaria baicalensis Georgi by studying the purification processes. Methods: The UV and HPLC were used as the analysis methods, and the contents and transfer rates of the total flavone, baicalin and wogonin were taken as the investigation indexes; the total flavone was purified by the precipitation separation with chlorhydric acid (PSCA) and HP20 macroporous adsorption resin; and the two methods were compared. Results: When the total flavone were purified by the PSCA, its degree of purity was 97.61%, the contents of the baicalin and wogonin were respectively, 9.19 g/100 g crude herb and 0.71 g/100 g crude herb, and the transfer rates were respectively 99.33%, 90.05%, and 82.96%; and when the total flavone were purified by the HP20 macroporous adsorption resin, its degree of purity was 80.66%, the contents of the baicalin and wogonin were respectively 8.97 g/100 g crude herb and 0.53 g/100 g crude herb, and the transfer rates were respectively 84.01%, 78.21% and 77.24%. Conclusion: The contents and transfer rates of the total flavone, baicalin, and wogonin dramatically increased when the PSCA was used, so the PSCA is the preferable technical process for the purification of the total flavone.
