左归丸对肾虚骨质疏松症大鼠ATF4与BGP表达的影响

| 摘 要： | 目的：通过研究左归丸对肾虚骨质疏松症大鼠血清中转录活化因子4（activating transcription factor-4，ATF4)与骨钙素（bone gla protein，BGP）蛋白表达的影响，探讨肾虚骨质疏松症的病理机制。方法：以摘除雌性大鼠双侧卵巢的方法复制肾虚骨质疏松症模型，采用具有补肾作用的左归丸对实验大鼠治疗12周，以盖天力片作为阳性对照组，正常大鼠、假手术组作为标准对照组，模型大鼠作为空白对照组（模空组），用ELISA法检测肾虚骨质疏松症大鼠血清ATF4，BGP的蛋白表达。结果：ELISA方法检测表明，正常大鼠血清中存在ATF4，BGP的蛋白表达。模空组血清中ATF4，BGP的蛋白表达水平明显下降。左归丸组、盖天力组用药12周后，与模空组相比，左归丸组可显著上调血清中ATF4，BGP的蛋白表达水平。结论：1） 正常大鼠血清中存在ATF4，BGP的蛋白表达；2） 骨质疏松症的发生可能与血清中ATF4，BGP的蛋白表达异常变化有关；3） 左归丸通过调控ATF4，BGP的蛋白表达，对骨质疏松症有一定的防治作用。 |

| Abstract： | Objective: To explore the etiological factors and pathologh of the osteoporosis due to kidney deficiency through studying the the effect of Zuogui pill on the expression of activating transcription factor-4 (ATF4) and bone gla protein (BGP) in blood serum of the osteoporosis rats. Methods: By using ovariectomy, we established the rat model of osteoporosis due to kidney deficiency. We treated the osteoporosis rat with Zuogui pills for 12 weeks. Simultaneously，the treatment with Gaitianli tablets as positive control groups. And rats in the normal group and model group were not given any treatment. We used the ELISA method to investigate the expression of the ATF4 and BGP in blood serum of the rats. Results: ELISA detection showed that the ATF4 and BGP existed in the normal blood serum. When comparing with the normal group and fake operational group，the level of the expression of the ATF4 and BGP in blood serum of model group decreased significantly. After the precaution treatment with Zuogui pills, the expression level of the ATF4 and BGP increased evidently in blood serum. Conclusions: 1) In normal rats, the blood serum can express the ATF4 and BGP in the protein level. 2) The changing of ATF4 and BGP protein expression in the blood serum may be one of the important occurring mechanism of the osteoporosis due to kidney deficiency. 3) The experimental Chinese herbs, which have the function of tonifying kidney, can increase the level of the expression of the protein of ATF4 and BGP in blood serum. This would prevent the osteoporosis resulted from kidney deficiency. |
