β-1,3-D-葡聚糖刺激免疫细胞表达相关细胞因子对体细胞的调控作用研究

摘 要: 目的:探究β-1,3-D-葡聚糖通过RAW264.7巨噬细胞细胞因子的表达对3T3成纤维细胞的增殖调控作用。方法:CCK-8检测β-1,3-D-葡聚糖水溶液对3T3成纤维细胞增殖的调控作用。RT-qPCR检测β-1,3-D-葡聚糖对RAW264.7巨噬细胞相关因子IL-1,IL-6和TNF-α表达的调控行为,并得到浓度剂量关系。冷冻干燥法合成β-1,3-D-葡聚糖支架,扫描电镜(scanning electron microscope,SEM)探究RAW264.7巨噬细胞在支架上的粘附和生长状况。通过共培养探究β-1,3-D-葡聚糖通过RAW264.7巨噬细胞对3T3成纤维细胞增殖的间接调控作用。结果:β-1,3-D-葡聚糖水溶液无法直接刺激3T3成纤维细胞增殖。β-1,3-D-葡聚糖可以上调巨噬细胞IL-1,IL-6和TNF-α的表达,并具有浓度依赖性。RAW264.7巨噬细胞可以粘附并生长于β-1,3-D-葡聚糖支架。接种RAW264.7巨噬细胞的β-1,3-D-葡聚糖支架与3T3成纤维细胞共培养可以刺激3T3成纤维细胞增殖,并具有时间依赖性。结论:β-1,3-D-葡聚糖支架可能通过调控RAW264.7巨噬细胞的相关因子表达刺激3T3成纤维细胞增殖。

Title: Research on β-1,3-D-glucan promoting somatic cells proliferation by activating cytokines expressions of immune cells

Abstract: Objective: The study is to investigate the effects of 餬-1,3-D-glucan on regulating 3T3 fibroblasts proliferation via the cytokines expressions of RAW264.7 macrophages. Methods: The soluble β-1,3-D-glucan’s effects on 3T3 fibroblasts proliferation were investigated by CCK-8 assay. The IL-1, IL-6 and TNF-α expressions of RAW264.7 macrophages treated with various concentrations of β-1,3-D-glucan were detected by RT-qPCR. β-1,3-D-glucan scaffold was fabricated by freeze-drying. The morphology of RAW264.7 macrophages seeded on the scaffolds was characterized by scanning electron microscope (SEM). 3T3 fibroblasts were co-cultured with β-1,3-D-glucan scaffold seeded with RAW264.7 macrophages, and 3T3 fibroblasts viability was investigated. Results: The soluble β-1,3-D-glucan does not have effects on 3T3 fibroblasts proliferation but it can promote RAW264.7 macrophages expressions of IL-1, IL-6 and TNF-α, and its immunomodulatory activities show dose-depended. RAW264.7 macrophages attach and spread on the scaffold. 3T3 fibroblasts proliferation is enhanced when they are co-cultured with β-1,3-D-glucan scaffold seeded with RAW264.7 macrophages, and their viabilities show time-depended. Conclusion: 餬-1,3-D-glucan can indirectly promote 3T3 fibroblasts proliferation through activation of RAW264.7 macrophages.
