RpoN和FlgM失活对大肠杆菌克拉酸合成的影响

摘 要: 本文针对大肠杆菌W3110菌株,利用CRISPR/Cas9基因编辑技术失活全局转录因子RpoN和FlgM,研究其对克拉酸(colanic acid,CA)合成的影响,并对发酵培养条件进行优化。结果显示,ΔrpoNΔflgM菌株的CA产量最高,在M9发酵培养基,28℃发酵条件下发酵瓶的CA产量最高达3 387.29 mg/L,比相同条件W3110产量提高了约234倍。本文显示转录因子RpoN和FlgM的共同失活对CA合成具有显著促进作用。

Title: Effect of RpoN and FlgM inactivation on colanic acid synthesis in Escherichia coli

Abstract: The paper targets Escherichia coli strain W3110 and uses CRISPR/Cas9 gene editing to inactivate the global transcription factors RpoN and FlgM, to investigate their effects on colanic acid (CA) synthesis and to optimize the culture conditions. The results showed that the CA production of ΔrpoNΔflgM strain was the highest, and that in M9 fermentation medium, the highest CA yield of fermentation bottles was up to 3 387.29 mg/L under the fermentation conditions of 28℃, which was about 234 times higher than the yield of W3110 under the same conditions. This thesis shows that the co-inactivation of transcription factors RpoN and FlgM has a significant contribution to CA synthesis.
