复方银黄微型灌肠剂对大鼠腹腔巨噬细胞NO及iNOS的影响

摘 要: | 研究复方银黄微型灌肠剂(Yinhuang micro-enema compound,YHMCE)对巨噬细胞释放一氧化氮(nitric oxide, NO)及诱导型一氧化氮合酶(induced nitricoxide synthase, iNOS)活性的影响,探讨其抗炎作用分子机理。体外培养大鼠腹腔巨噬细胞(peritoneal macrophage,PMФ),采用氧化还原酶法、免疫细胞化学法和RT-PCR分别测定不同剂量YHMCE对内毒素(lipopolysaccharide, LPS)诱导大鼠腹腔巨噬细胞NO生成量、iNOS活性及iNOS mRNA表达水平的影响。结果表明:YHMCE能降低大鼠腹腔巨噬细胞iNOS的活性及iNOS mRNA的表达水平,减少LPS诱导的NO大量产生和释放,并呈作用剂量依赖关系。YHMCE对大鼠腹腔巨噬细胞NO产生的调节可发生在iNOS的转录水平,抑制iNOS从头合成,减少NO合成与释放,可能是其发挥抗炎作用的分子作用机理之一。 |

Abstract: | To study the effect of Yinhuang micro-enema compound (YHMCE) on release of nitric oxide (NO) and the activity of induced nitric oxide synthase (iNOS) so as to elucidate its anti-inflammatory mechanisms. Peritoneal macrophage (PMФ) obtained from normal rats were cultivated in vitro. The production of NO, the activity and mRNA expression of iNOS in peritoneal macrophages of mice administered with different dose of YHMCE were observed by nitrate reductase method, immunocytochemical method and RT-PCR, respectively. YHMCE can inhibit the activity and mRNA expression of iNOS with dose-dependence in rat peritoneal macrophages, and reduce the generation and release of NO. The regulation of YHMCE on the production of NO in peritoneal macrophages of mice may occur at transcriptional level of iNOS, inhibiting the iNOS’s denovo synthesis and reducing the generation and release of NO. This may be one of its anti-inflammatory mechanisms. |
