Vx2肝转移癌动物模型传代方法的比较

摘 要: | 目的:探讨高成瘤率兔Vx2移植瘤后肢肌肉传代与肝脏传代在兔存活时间及成瘤质量等方面的不同。方法:采用瘤块移植法建立兔Vx2后肢肌肉模型及肝癌模型,采用磁共振技术检测肿瘤生长情况,定期在兔后肢肌肉和肝脏的移植部位作瘤组织活检,了解肿瘤生长情况,记录并比较实验兔的生长情况和存活时间。结果:采用瘤块移植方法成功建立了稳定的兔Vx2后肢肌肉及肝脏肿瘤模型,后肢移植成瘤情况为20/25,肝移植成瘤情况为22/25,2组的成瘤率无显著性差别(P>0.05)。后肢组与肝脏组在移植第7天和第14天后的肿瘤体积大小比较:后者显著大于前者(P<0.01)。接种后第10天的肿瘤生长率(tumor growth rat, TGR)前者为64.9%,后者为94.6%;接种后第20天的TGR前者为70.1%,后者为96.7%. 荷后肢移植瘤兔存活时间达到30 d的只数为3/20,荷肝脏移植瘤兔为20/22,比较2组荷瘤兔存活时间,肝脏组明显长于后肢组(P<0.05)。结论:肝脏移植瘤体积大、实质细胞丰富、肿瘤坏死情况少、生存期长,是比较理想的传代模型。 |

Abstract: | Objective: To investigate the difference in rabbits’ survival time and transplanted tumor quality between two transplanted groups. One group is the rabbit Vx2 transplantation tumor transplanted into rabbits’ muscle of posterior limb for going down to the future generation, the other is into rabbits’ liver. As known the rabbit Vx2 transplantation tumor’s growth rate is high. Methods: Employ tumor transplants to set up the muscle tumor model and the liver cancer model from rabbit Vx2 tumor. Apply magnetic resonance to detect the tumorous growth. Biopsy the tumor tissues which grow in rabbits’ muscle of posterior limb or in liver regularly, in order to get the message about tumorous growth. Record and compare the experimental rabbits’ tumorous growth and the rabbits’ survival time. Results: Set up stable rabbit Vx2 posterior limb’s muscle tumor model and liver tumor model successfully by tumor transplants, within the 25 rabbits’ muscle of posterior limb being transplanted, about 20 had tumor growth ultimately, the other 25 rabbits were transplanted into liver, about 22 had tumor growth ultimately, the tumor formation rate of the two groups was alike (P>0.05). Comparison of tumors’ volume at the 7th day and the 14th day after transplantation between the posterior limb group and the liver group showed that the latter exceeded the former significantly (P<0.01). The tumor’s TGR of the former group was 64.9% at the 10th day after transplantation, the latter group was 94.6%; then the former was 70.1% and the latter was 96.7% at the 20th day after transplantation. About 3/20 rabbits with tumor in the muscle of posterior limb and 20/22 in the liver could survive for 30 days(P<0.05). Conclusion: Contrast to the muscle tumor group, the tumor volume of liver cancer group is larger, and its growing period is longer, the tumor parenchymal cells are richer, and cellular necrosis degree is smaller, which indicates that it is a relatively ideal passage model. |
