初生乌骨鸡与蛋、肉鸡肝脏组织基因转录阶段的差异探索

摘 要: 目的:探索中国珍贵的家禽品种资源——丝羽乌骨鸡和四川山地乌骨鸡特异性状相关蛋白的表达和调控机制。方法:以蛋鸡和肉鸡为对照,结合银染差异显示逆转录聚合酶链式反应(differential display reverse transcription polymerase chain reaction,DDRT-PCR)技术和基因芯片技术筛选1日龄乌骨鸡肝脏组织差异显示的表达序列标签(expressed sequence tag, EST)。结果:筛选到3条真实的差异表达cDNA序列。DQ003271仅在蛋鸡中表达,为家鸡新的EST,位于5号染色体;DQ003272仅在乌骨鸡中表达,为家鸡已知EST,属于Gga.9136基因簇,contig 1的正向阅读框1序列与NP_001012582.1一致性达91%,推测该片断所在的基因极可能直接影响酪氨酸酶的表达,控制“乌色”性状;DQ003274未在山地乌骨鸡中表达,与鸡肠淋巴细胞cDNA克隆序列CD737238.1一致性达100%,属基因簇Gga.8993.结论:为进一步研究乌骨鸡的独特性状及乌骨鸡、蛋鸡、肉鸡三者间的差异性状提供了重要信息。

Title: Study on genetic transcription difference of liver tissues of nascent black-bone chicken, layer and broiler

Abstract: Objective: To study expression and regulation mechanisms of proteins related with specific characteristics in Silky fowl and Sichuan mountain black-bone chicken. Methods: Differentially display reverse transcription polymerase chain reaction (PCR) and gene chip scanning were adopted to attain expressed sequence tag (EST) which were differentially displayed among the black-bone chicken, the layer and the broiler. Results: Three ESTs were obtained. Bioinformation analysis proved that DQ003271 only expressed in the layer was a new EST, located at the fifth chromosome; and DQ003272 only expressed in the black-bone chicken was a known EST, belonging to Gga.9136. The first sequence of positive reading frame of contig 1 was 91% homologous to NP_001012582.1. The gene including the segment may directly control the expression of Tyrosinase, thereby controlling the black color trait. DQ003274 only not expressed in the Sichuan mountain black-bone chicken had 100% consistency with CD737238.1 and belonged to Gga.8993. Conclusion: These results provide significant data for further research on different traits among the black-bone chicken, the layer and the broiler.
