番茄microRNA及其靶基因对几种病毒侵染的反应

摘 要: microRNA(miRNA)是植物中一个重要基因调控机制——miRNA基因沉默途径的重要组成部分。实验选用不同致病能力的黄瓜花叶病毒株系和番茄不孕病毒(CMV-Fny, CMV-FnyΔ2b, TAV-Bj)侵染番茄,利用stem-loop荧光定量反转录聚合酶链反应(real-time reverse transcription polymerase chain reaction, real-time RT-PCR),分析了不同侵染时期、不同寄主组织中的7条miRNA及其部分靶标mRNA对病毒侵染的响应,探讨了不同病毒与寄主互作过程中寄主miRNA调控途径的改变和影响。这其中所表现出的现象和规律与病毒对寄主防御机制的破坏、miRNA及其靶标mRNA的时间-空间特异性表达、特定miRNA的生物学调控功能等有着密切联系。

Title: Response of tomato microRNA and their target mRNAs to virus infection

Abstract: MicroRNA (miRNA) plays an important role in miRNA gene silencing pathway, which contributes a lot in plant developmental system. This experiment chose three plant viruses from different strains (CMV-Fny, CMV-FnyΔ2b, TAV-Bj) and inoculated them to tomato seedlings. At different periods of infection, the expressing levels of seven miRNAs and partial mRNAs were quantified in four different kinds of tissues by stem-loop real-time reverse transcription polymerase chain reaction (real-time RT-PCR). The analysis studied the change and influence of miRNA gene regulatory pathway in plant bodies during the interaction between plant virus and their hosts. The exhibited phenomena and laws have significant relationship with the damage to the host defense mechanism caused by virus, time-space specificity expression of the miRNA and the target mRNA, biological regulation function of the specific miRNA, and so on.
