用木瓜蛋白酶和L-半胱氨酸建立大鼠膝关节骨关节炎模型的研究

摘 要: 目的:探讨采用木瓜蛋白酶和L-半胱氨酸在大鼠膝关节腔内注射,建立大鼠骨关节炎(osteoarthritis, OA)动物模型,为研究该疾病的康复干预奠定基础。方法:对20只SD(Sparague-Dawley)大鼠行膝关节腔内注射,实验侧分别在第1,3,7天注射4%的木瓜蛋白酶溶液与0.03 mol/L的L-半胱氨酸溶液混合液20 μL,对照侧注射等量生理盐水,首次注射后第1,2,4周分批测量大鼠膝关节活动范围并处死实验动物,切取膝关节软骨做大体观察及组织病理学观察。结果:实验侧关节注射木瓜蛋白酶后,活动范围明显低于对照侧,关节软骨变薄且有裂隙形成、纤维变性、基质分解破坏等退行性改变,并随观察时间的延长而进一步发展。对照组并无类似改变,两者比较差异有显著性意义。结论:大鼠膝关节腔内注射木瓜蛋白酶和L-半胱氨酸后的改变与人膝关节骨关节炎病理改变具有相似性,通过该模型可以研究膝关节骨性关节炎(knee joint osteoarthritis,KOA)病理、发病机理及治疗方法。

Title: A study on papain and L-cysteine induced osteoarthritis in rat knee joint

Abstract: Objective: To establish an animal model of osteoarthritis (OA) in rat knee joint so as to provide a basis for the rehabilitative intervention of OA. Methods:20 μL mixture of 4% papain solution and 0.03 mol/L L-cysteine solution was injected into articular cavity of 20 Sparague-Dawley (SD) rats on the first, third and seventh day to establish the model of knee osteoarthritis and the same volume of saline was injected into the contralateral articular cavity as control. The activity scope of knee joint was measured in batches on the first, second, fourth week after the first injection and the rats were sacrificed and the knee joint cartilages were cut for general observation and histopathological examination. Results: The degenerative changes were demonstrated in the knee joints injected with papain and L-cysteine, such as the lower activity scope of knee joint, thinner and worse articular cartilage, fissuration, fibrous degeneration, fibrillation and destroyed cartilage matrix. All these changes were much worse with prolonged observation time. No similar changes were found in the control group, with significant difference between them. Conclusion: Pathological similarities between papain induced rats knee osteoarthritis and human knee osteoarthritis were observed. This animal model can be used for exploring pathogenesis of this disease and selection of treatments for osteoarthritis of knee joint.
