黄芪多糖对慢性粒细胞白血病患者浆细胞样树突状细胞功能的影响

| 摘 要： | 目的：通过体外实验研究黄芪多糖（Astragalus polysaccharide，APS）对慢性粒细胞白血病（chronic myelocytic leukemia，CML）患者浆细胞样树突状细胞（plasmacytoid dendritic cells，pDC）功能的影响，为进一步阐明APS的免疫学活性及其在免疫治疗中的应用提供依据。方法：从慢性粒细胞白血病患者外周血分离单个核细胞，免疫磁珠法去除pDC，加入0，50，100，200 mg/L的APS共孵育24 h，收集上清液，应用酶联免疫吸附实验检测pDC分泌的IFN-α，IL-6，TNF-α量的变化。结果：实验应用慢性粒细胞白血病患者外周血来源的pDC，通过体外实验证明了APS能够提高治疗后的慢性期CML患者pDC分泌的IFN-α，IL-6，TNF-α量及初诊未治慢性期CML患者pDC分泌的IL-6，TNF-α量，并且呈APS浓度依赖性。结论：实验结果证实了APS能够增强慢性粒细胞白血病患者pDC的功能。 |

| Abstract： | Objective: To explore the mechanism of immunomodulatory activity of Astragalus polysaccharide (APS) on chronic myelocytic leukemia (CML) peripheral blood mononuclear cells (PBMC)-derived plasmacytoid dendritic cells (pDC) by in vitro experiment, and further explain immunomodulatory activity of APS and its application in immunotherapy. Methods: The experiment isolated chronic myelocytic leukemia peripheral blood mononuclear cell (PBMC) and isolated pDC by magnetic activated cell sorting (MACS), then incubated with APS (0, 50, 100, 200 mg/L) for 24 h, and used enzyme-linked immuno sorbent assay (ELISA) to determine the variation of IFN-α, IL-6, TNF-α excreted by pDC in supernate. Results: The experiment used pDC of peripheral blood of chronic myeloid leukemia (CML) patients, and proved by in vitro experiment that the APS could increase the concentration of IFN-α, IL-6, TNF-α secreted by pDC of the treated CML patients and the concentration of IL-6 and TNF-α secreted by pDC of the untreated CML patients, which depended on the APS concentration. Conclusion: APS can promote the function of pDC from CML. |
