芪芍五味子颗粒对病毒性心肌炎小鼠心肌细胞凋亡和氧化应激状态的影响

摘 要: 目的:研究病毒性心肌炎(viral myocarditis,VM)模型小鼠心肌细胞凋亡和氧化应激状态及中药芪芍五味子颗粒对其的影响。方法:BALB/c小鼠接种柯萨奇B3病毒(coxsackie virus B3,CVB3),建立病毒性心肌炎动物模型,随机分为病毒对照组、芪芍五味子颗粒中药治疗组及VitC和病毒唑联用治疗组。各组于实验第3,7,14,21天分批处死,取心脏用流式细胞仪检测心肌细胞凋亡和坏死率,并作组织病理学和电镜检查,同时分别用黄嘌呤氧化酶法和硫巴比妥酸法测血清超氧化物岐化酶(superoxide dismutase,SOD)活性及丙二醛(maleic dialdehyde,MDA)含量。结果:VM小鼠心肌细胞凋亡率和坏死率均显著增加,凋亡高峰在CVB3感染后第7天,坏死高峰在第14天,与组织形态学观察一致,与病毒对照组比较,中药治疗组小鼠心肌细胞凋亡率和坏死率显著降低(P<0.05),小鼠心肌炎症性病理变化明显减轻;病毒对照组小鼠处于氧化应激状态,中药组小鼠在第14,21天血清SOD活性明显高于病毒对照组,MDA含量明显低于病毒对照组(P<0.05)。结论:芪芍五味子颗粒能减轻CVB3感染小鼠心肌病理损害程度,调整其氧化应激状态,降低心肌细胞凋亡率和坏死率,对VM有良好的防治作用。

Title: Qishaowuweizi compound�s regulating effect on cardiomyocyte apoptosis and oxidative stress of viral myocarditis model mice

Abstract: Objective: To investigate cardiomyocyte apoptosis and oxidative stress in viral myocarditis (VM) model mice and treatment effect of Qishaowuweizi compound. Methods: Male BALB/c mice were infected intraperitoneally with coxsackie virus B3 (CVB3) to prepare VM model and randomly divided into normal control group, virus control group, Qishaowuweizi compound treatment group and ribavirin plus VitC treatment group. Mice were sacrificed on experiment day 3, 7, 14 and 21 postinoculation (p.i.). Cardiomyocyte apoptosis and necrosis were detected by flowcytometry (FCM). Histomorphologic changes were observed under light microscope and electron microscope. Besides, research was done on serum superoxide dismutase (SOD) activity by xanthine oxydase method and maleic dialdehyde (MDA) by thiobarbituric acid (TBA) method. Results: In virus control group, cardiomyocyte apoptosis and necrosis both increased compared with normal control group (P<0.05). The apoptosis peak appeared on day 7 p.i. and the necrosis peak on day 14 p.i., consistent with histomorphologic observation. While compared with virus control group, cardiomyocyte necrosis were lower in Qishaowuweizi compound treatment group; the pathologic changes were meliorated. Mice of virus control group were in oxidative stress condition. In Qishaowuweizi compound treatment group, the serum SOD was more active and the value of MDA was less than virus control group on day 14 p.i. and 21 p.i (P<0.05). Conclusion: Qishaowuweizi compound could meliorate cardiac pathologic changes of VM mice, regulate their oxidative stress condition and reduce cardiomyocyte apoptosis and necrosis, having predictably favorable effect in treating VM.
