化合物茯苓酸对鼠外周血淋巴细胞线粒体跨膜电位的影响

摘 要: 目的:研究茯苓酸在活体动物中是否可以调节鼠外周血淋巴细胞线粒体跨膜电位,确定该跨膜电位是否是茯苓酸的药理学靶点之一。方法:SD大鼠分别予以茯苓酸1.0,10.0(mg/kg)·d-1,环孢素A(CsA)5.0(mg/kg)·d-1和同体积橄榄油灌胃,7 d后通过流式细胞仪检测其外周血淋巴细胞线粒体跨膜电位变化的细胞百分比。结果:数据提示,茯苓酸可以稳定外周血淋巴细胞线粒体跨膜电位,避免其去极化,而且相对于较小剂量茯苓酸[1.0(mg/kg)·d-1]而言,较大剂量的茯苓酸[10.0(mg/kg)·d-1]表现出更好的药理学作用。结论:口服茯苓酸能稳定细胞线粒体跨膜电位,减少其去极化百分比,这与体外研究结果不同。

Title: Effect of compound pachymic acid on peripheral blood lymphocytes mitochondrial transmembrane potential in rats in vivo

Abstract: Objective: To study the effect of pachymic acid (PA), a natural triterpenoid, to peripheral blood lymphocytes (PBLs) mitochondrial transmembrane potential in rats. Methods: In this study, PBLs mitochondrial transmembrane potential were measured in rats 7 days after intragastric administration using different dose PA [1.0, 10.0 (mg/kg)·d-1], CsA[5.0 (mg/kg) ·d-1] and olive oil as control group. Results: Data suggested that PA treatment protect PBLs mitochondrial transmembrane potential from depolarization. Large dose of PA [10.0(mg/kg)·d-1]has better pharmacological effect than small dose of PA [1.0 (mg/kg)·d-1] .Conclusion: The findings indicate that PA has a protective effect acting to PBLs through a novel mechanism involving stabilization of the PBLs mitochondrial transmembrane potential.
