肠道病毒71型重症患儿分离株SDLY96的全基因组序列分析

| 摘 要： | 目的：对肠道病毒71型（Enterovirus 71，EV71）重症患儿分离株SDLY96进行全基因组序列测定与分析，为构建EV71感染性克隆奠定基础。方法：利用RT-PCR对2010年山东临沂1例手足口病(hand-foot-and-mouth disease, HFMD)重症患儿标本分离到的EV71型分离株(SDLY96)进行全基因组核苷酸序列测定,并对基因组序列进行同源性比较及遗传进化分析。结果：SDLY96株基因组全长7 405个核苷酸，SDLY96株与Fuyang株EU703813的核苷酸及氨基酸序列同源性均最高,整个基因组核苷酸同源性为98.4%,氨基酸同源性为99.1%. 基于VP1区的基因遗传进化分析显示，SDLY96株与Fuyang株EU703813进化关系最为接近。结论：SDLY96株属EV71病毒的C4基因亚型的C4a分支，与Fuyang株的核苷酸及氨基酸序列同源性很高。 |

| Abstract： | Objective: To analyze the complete genome sequence of an Enterovirus 71 (EV71) strain SDLY96 isolated from a severe case and lay the foundation for constructing infectious clone of EV71. Methods: The complete genomes of SDLY96 from a sufferer of hand-foot-and-mouth disease (HFMD) in Linyi city of Shandong province were amplified by RT-PCR. The homology of nucleotide and amino acid sequences were aligned, and phylogenetic relations of the sequences were analyzed. Results: The full length of SDLY96 strains was 7 405 bp. SDLY96 strain shared the high homology with Fuyang strain EU703813 both in nucleotide and in amino acid sequences, with 98.4% identity in nucleotides and 99.1% in amino acids. The full VP1 region based phylogenetic analysis also indicated that strain SDLY96 had the close genetic relationship with Fuyang strain EU703813. Conclusion: SDLY96 should be classified as C4a branch of subgenogroup C4 of EV71, and SDLY96 strain has close genetic relationship with Fuyang strain. |
