板蓝根抗病毒有效部位中的化学成分

摘 要: 目的:研究板蓝根抗病毒有效部位中化学成分的结构和体外抗病毒活性。方法:板蓝根水提物经大孔树脂吸附,10%,50%(体积比)乙醇洗脱得抗病毒有效部位,经硅胶柱色谱、反相硅胶色谱及Sephadex LH-20反复分离纯化得到单体,测定所得化合物的理化常数和波谱数据,鉴定其化学结构。噻唑蓝(methyl-thiazolyldiphenyl-tetrazolium,MTT)检测所得单体化合物体外对单纯疱疹病毒(herpes simplex virus,HSV)有抑制作用。结果:从板蓝根水提抗病毒有效部位中分离得到10个化合物,分别鉴定为丁香苷(1)、4-(1,2,3-三羟基丙基)-2,6-二甲氧基苯-1-O-β-D-葡萄糖苷(2)、异落叶松脂醇(3)、异落叶松脂醇-4-O-β-D-葡萄糖苷(4)、落叶松脂素-4-O-β-D-葡萄糖苷(5)、落叶松脂素-4,4′-二-O-β-D-二葡萄糖苷(6)、2-羟基-1,4-苯二甲酸(7)、D-甘露醇(8)、吲哚-3-乙腈-6-O-β-D-葡萄糖苷(9)、3-[2′-(5′-羟甲基)呋喃基]-1(2H)-异喹啉酮-7-O-β-D-葡萄糖苷(10)。结论:其中1个化合物(化合物10)为新化合物,4个化合物为首次从板蓝根药材中分离得到。化合物10具有明显的抗HSV-2型病毒作用,药物浓度为0.125 g/L时病毒抑制率为57.31%,半数有效量(50% concentration of inhibition,IC50)为0.093 g/L,治疗指数(therapeutic index,TI)为7.114.

Title: Chemical composition from antivirus active parts of Radix Isatidis

Abstract: Objective: To research the the structure and chemical composition of antiviral effective activity part of Radix Isatidis. Methods: The aqueous extract of Radix Isatidis was absorbed by macroporous resin, the effective antiviral parts were obtained through 10%, 50% ethanol, and the purified monomer was obtained by silica gel column chromatography, reversed phase chromatography on silica gel and Sephadex LH-20 repeatedly separation. Then their physico-chemical constants and spectral data were evaluated and their chemical structures were identified. The inhibitory effect on herpes simplex virus (HSV) of resulting compounds were detected by methyl-thiazolyldiphenyl-tetrazolium (MTT) assay in vitro. Results: Ten compounds were isolated from the active part of water extract, they were respectively identified as syringin (1), 4-(1,2,3-trihydroxypropyl)-2,6-dimethoxyphenyl-1-O-β-D-glucopyranoside (2), isolariciresino (3), isolariciresinol-4-O-β-D-glucopyranoside (4), lariciresinol-4-O-β-D-glucopyranoside (5), lariciresinol-4,4′-bis-O-β-D-glucopyranoside (6), 2-hydroxy-1,4-benzenedi carboxylic acid (7), D-mannitol (8), Indole-3-acetonitrile6-O-β-D-glucopyranoside (9), 3-[2′-(5′-hydroxymethyl)furyl]-1(2H)-isoquinoline-7-O-β-D- glucopyranoside (10). Conclusion: One of these compounds was a new compound, and four were isolated from Radix Isatidis at the first time. The new compound 10 had obvious effects against HSV-2. When the drug concentration was 0.125 g/L, the viral suppression was 57.31%, 50% concentration of inhibition (IC50) was 0.093 g/L, and therapeutic index (TI) was 7.114.
