绿色荧光蛋白转基因昆明白小鼠的建立

摘 要: 目的:建立表达绿色荧光蛋白(green fluorescence protein,GFP)的转基因昆明白小鼠,为相关实验提供携带报告基因的远交系动物模型。方法:用携带有GFP报告基因的慢病毒载体感染昆明小鼠胚胎干细胞(embryonic stem cell, ESC)系KES,得到稳定高效表达GFP的ESC KES-GFP. 通过将KES-GFP与129品系小鼠的胚胎进行聚合发育形成嵌合体囊胚,移植到假孕小鼠子宫获得表达GFP的嵌合体小鼠,嵌合体小鼠与昆明白小鼠交配获得转基因小鼠。结果:成功获得了表达GFP的转基因昆明白小鼠。结论:基于ESCs成功建立了GFP转基因昆明白小鼠。

Abstract: Objective: To establish transgenic Kunming white mice expressing green fluorescence protein (GFP), provide outbred animal models carrying reporter gene for related studies. Methods: Using infected Kunming mouse embryonic stem cell line KES with a lentivirus carrying GFP as a reporter gene, an embryonic stem cell (ESC) line KES-GFP expressing GFP was generated efficiently and stably. These KES-GFP cells were introduced into host embryos from 129 stain mouse through aggregation to form chimeric blastocysts, then transferred it into uterus of pseudopregnant mice to obtain chimeric mice expressing GFP. These chimeric mice were mated with Kunming mice to obtain transgenic mice. Results: We obtained transgenic Kunming white mice expressing GFP successfully. Conclusion: Transgenic Kunming white mice expressing GFP can be successfully established based on ESCs.
