家蚕JAK-STAT信号通路调控的基因预测

摘 要: 研究通过生物信息学分析预测了Janus激酶信号转导和转录激活子(Janus kinase/signal transducer and activator of transcription,JAK-STAT)信号通路调控的基因。从家蚕基因组数据库中下载所有预测基因,通过扫描搜索启动子区域(1 000 bp)的STAT结合序列TTCN2-4GAA发现:家蚕的9 293个基因编码7 271 个非冗余蛋白,其基因的启动子序列中存在1~6个靶序列。基因本体论(gene ontology,GO)注释的结果显示:这些蛋白质参与细胞组成、分子功能和生物学过程,表明JAK-STAT通路对很多基因发挥调节作用。靶基因中超过50%的基因与结合相关,约40%的基因与细胞过程、代谢过程和催化活性相关。通常认为JAK-STAT 通路在免疫反应中也发挥作用,在一些与抗病毒因子、细胞免疫效应和小分子抗菌肽(如防卫素、攻击素、moricin和葛佬素蛋白3)等相关基因的启动子区域可以检测到1~4个STAT结合靶序列,暗示一些抗病毒因子、细胞免疫效应物和抗病毒活性相关的抗菌肽表达可能由JAK-STAT信号通路调控。

Title: Prediction of genes regulated by JAK-STAT signal pathway of Bombyx mori

Abstract: In the present study, the genes regulated by Janus kinase/signal transducer and activator of transcription pathway (JAK-STAT) signal pathway were predicted by bioinformatics analysis. 1 000 bp of upstream promoter sequence of the all predicted genes were downloaded from the silkworm genome database, and the STAT binding sequence TTCN2-4GAA was searched by scanning the promoter sequences. The results showed that 1-6 the target sequences could be found in the upstream promoter sequences of 9 293 genes coding 7 271 non-redundant proteins. Gene ontology (GO) annotation results showed that these proteins were involved in cellular component, molecular function and biological process, suggesting JAK-STAT pathway played an important role in many ways. More than 50% genes were related to binding, about 40% genes were related to cellular process, metabolic process and catalytic activity in the targeting genes. It is considered that JAK-STAT plays a role in immune response, 1-4 STAT binding sequences could be detected in promoter region of some genes related to anti-viral factors, cellular immune effector, and small antimicrobial peptide such as defensin, attacin, moricin and gloverin3, implying that the expression of some anti-viral factors, cellular immune effectors and antimicrobial peptides related to antiviral activity might be regulated by JAK-STAT signal pathway.
