补肾阳法对卵巢切除大鼠血清褪黑素水平和骨组织代谢的影响

摘 要: | 目的:观察补肾阳法对卵巢切除(ovariectomized,OVX)所致骨质疏松大鼠血清褪黑素(melatonin,MT)水平和骨组织代谢产物骨组织特异性碱性磷酸酶(bone alkaline phosphotase,B-ALP)、Ⅰ型前胶原氨基末端前肽(procollagen typeⅠamino-terminal propeptide,PINP)、Ⅰ型胶原氨基末端肽(N-telopeptide of typeⅠcollagen,NTX)和Ⅰ型胶原羧基末端肽(C-telopeptide of typeⅠcollagen,CTX)的影响,探讨补肾阳法治疗绝经后骨质疏松的可能机制。方法:24只雌性SD大鼠随机分为3组,假手术组、模型组和补肾阳组。卵巢切除术后3个月,补肾阳组予桂附地黄丸0.92 g/(kg·d)灌胃干预,其余2组予等量生理盐水灌胃。给药 3个月后采用双能X线(dual-energy X-ray absorptiometry,DXA)骨密度(bone mineral density,BMD)仪测定大鼠右侧股骨BMD,HE染色观察左侧股骨骨组织形态学,ELISA检测血清B-ALP,PINP,NTX和CTX水平,高效液相色谱法(high perfermance liquid chromatography,HPLC)测量血清MT含量。结果:补肾阳法可明显提高大鼠BMD,改善骨组织微观结构,降低血清B-ALP,PINP,NTX和CTX水平,提高血清MT水平。结论:补肾阳法可有效治疗大鼠卵巢切除所致的骨质疏松症,其作用机制与降低骨转换率并提高血清MT水平相关。 |

Abstract: | Objective: To observe the effect of tonifying kidney yang on serum melatonin (MT), bone alkaline phosphotase (B-ALP), procollagen typeⅠ amino-terminal propeptide (PINP), N-telopeptide of typeⅠcollagen (NTX) and C-telopeptide of typeⅠcollagen (CTX) in ovariectomized (OVX) rat, explore possible mechanism of its efficacy on postmenopausal osteoporosis. Methods: 24 SD rats were divided into three groups, sham operation group, OVX group and tonifying kidney yang group. Three months after the operation, rats in tonifying kidney yang group were given intragastric administration with Guifudihuang pill by 0.92 g/(kg·d), and rats in the rest two groups were given intragastric administration with saline. Three months after medicine intervention, bone mineral density (BMD) of right femur was detected with dual-energy X-ray absorptiometry (DXA), the bone morphology of left femur was observed with HE staining, serum levels of B-ALP, PINP, NTX and CTX were detected with ELISA, and serum level of melatonin were detected with high performance liquid chromatography (HPLC). Results: Tonifying kidney yang can significantly increase BMD, ameliorate bone microstructure, decrease serum levels of B-ALP, PINP, NTX and CTX, and improve serum level of MT. Conclusion: Tonifying kidney yang has efficacy to treat OVX-induced osteoporosis in rat, reducing bone turnover rate and improving serum level of MT might be the mechanism. |
