HPLC同时测定三黄片中盐酸小檗碱、黄芩苷和大黄素的含量

| 摘 要： | 目的：建立利用高效液相色谱（high performance liquid chromatography，HPLC）法同时测定盐酸小檗碱、黄芩苷和大黄素含量的方法并用于测定三黄片中三种主要药效成分的含量。方法：采用Agilent C18（4.6 mm × 250 mm，5 μm）色谱柱，流动相为乙腈-0.5%磷酸溶液（三乙胺调pH=4.0），梯度洗脱，流速1 mL•min-1，检测波长275 nm. 结果：盐酸小檗碱、黄芩苷的线性范围为1～30 μg∙mL-1，大黄素的线性范围为0.5～15 μg∙mL-1，方法回收率为95.83%～100.09%. 结论：该方法简便、准确、重复性好，定量限低并且能排除其他成分的干扰，可用于三黄片质量控制的评价。 |

| Abstract： | Objective: A rapid and specific high performance liquid chromatography (HPLC) method was developed for the analysis of berberine hydrochloride, baicalin and emodin. The method was applied to study the content determination of Sanhuang tablets containing berberine hydrochloride, baicalin and emodin. Methods: The analysis was carried out on a Agilent C18 reversed-phase column (4.6 mm × 250 mm, 5 μm) by gradient elution with acetonitrile and 0.5% phosphoric acid (adjusted pH=4.0 with triethylamine). The flow rate was 1 mL•min-1 and the detection wavelength was set at 275 nm. Results: Linear responses were obtained for berberine hydrochloride and baicalin ranging from 1 to 30 μg∙mL-1, emodin ranging from 0.5 to 15 μg∙mL-1. The extraction recovery ranged from 95.83% to 100.09%. Conclusion: This method is simple, accurate with good repeatability, low detection limit and can eliminate the interference of other components. This validated method could be used in the quality control of Sanhuang tablets. |
