不同癌症中PC的表达差异与临床意义研究

摘 要: 目的:探究丙酮酸羧化酶(pyruvate carboxylase,PC)在临床不同类型肿瘤样本及多种癌细胞系中的表达差异并分析其潜在意义。方法:通过实时荧光定量PCR(quantitative real-time PCR,qRT-PCR)与Western blotting等实验方法检测PC在临床肿瘤样本与癌细胞系中mRNA和蛋白的表达水平差异;利用shRNA沉默肝癌细胞Hep3B、肺癌细胞A549中的PC基因并初步检测其对细胞增殖的影响。结果:PC的mRNA水平与蛋白水平在临床肝癌样品中整体呈下降趋势,但是在临床肺癌样品中呈上升趋势;PC在肝癌细胞系中表达量相对较低,在肺癌细胞系中表达量相对较高。结论:PC在不同临床肿瘤样本及多种癌细胞系中的表达量有差异,具体表现为在肝癌中表达量下降而在肺癌中表达量上升。

Title: Study of expression difference and clinical significance of pyruvate carboxylase in different cancers

Abstract: Objective: To explore the expression difference and potential significance of pyruvate carboxylase (PC) in clinical tumor samples and various cancer cell lines. Methods: Quantitative real-time PCR (qRT-PCR) and Western blotting are used to detect the mRNA and protein levels of PC in clinical tumor samples and corresponding cancer cell lines. shRNAs are used to silence PC gene expression in hepatoma cell Hep3B and lung cancer cell A549. Results: The expression of mRNA level and protein level of PC is entirety decreased in clinical liver cancer samples compared with corresponding para-carcinoma tissues, whereas the opposite phenomenon is observed in clinical lung cancer samples compared with control tissues. Moreover, low PC protein level is found in hepatocellular carcinoma cell line compared with other cancer cell lines including lung cancer cells. Conclusion: PC expression is downregulated in hepatocellular carcinoma, whereas upregulated in lung carcinoma both at mRNA level and protein level, respectively.
