慢性高眼压大鼠动物模型的视网膜和视网膜神经节细胞变化研究

摘 要: 目的:观察高眼压对大鼠视网膜和视网膜神经节细胞(retina ganglion cell,RGC)的损伤。方法:采用巩膜上静脉烙闭(episcleral vein cauterization,EVC)的方法建立慢性高眼压模型,用Goldmann鼠眼压计在造模前测定基础眼压。造模后测量各组大鼠第l,3,5,7,28,56天的眼压,检测大鼠RGC数量,测量视网膜内丛状层的厚度。结果:巩膜上静脉烙闭的方法能成功建立慢性高眼压模型。高眼压状态下,大鼠RGC的数量明显变少,并且发生空泡样变,视网膜内丛状层的厚度也明显变薄。结论:慢性高眼压状态下,大鼠的视网膜变薄,RGC明显变少并凋亡。

Title: Study of retina and retina ganglion cells variation in a rat model of chronic ocular hypertension

Abstract: Objective: To investigate the effect of intraocular pressure (IOP) on retina and retina ganglion cell (RGC) in a rat model of chronic ocular hypertension. Methods: Chronic ocular hypertension was induced by episcleral vein cauterization (EVC) in rats. IOP was measured by Goldmann rat tonometer before operation and at 1, 3, 5, 7, 28, 56 days after operation. Retinal histologic sections were measured to determine inner plexiform layer (IPL) thickness and the number of RGC. Results: The model of chronic ocular hypertension was successfully established by EVC. High IOP significantly decreased the number of RGC and the thickness of retina IPL, and induced the vacuolation. Conclusion: High IOP can significantly decrease the thickness of retina and the number of RGC, and may cause the apoptosis in a rat model of chronic ocular hypertension.
