结核分枝杆菌铁离子依赖调控蛋白Fur和DtxR的研究进展

摘 要: | 对于包括结核分枝杆菌在内的大多数生物而言,铁既是一种必需元素,又具有潜在危险性。它作为酶的辅因子,参与重要的细胞功能,从细胞呼吸到DNA复制均必不可少。同时,铁离子能够催化芬顿反应的进行,导致有氧代谢过程中产生有毒的活性氧自由基,因而具有潜在的危险性。本文主要概述细菌中铁离子依赖的调控作用以及结核分枝杆菌铁离子调控蛋白Fur家族和DtxR家族的生物学功能,探究铁离子代谢对结核分枝杆菌的调控机制,研究将为结核病的治疗提供新方法。 |

Abstract: | To most living organisms, including Mycobacterium tuberculosis (Mtb), iron is an essential but potentially harmful nutrient. As a cofactor of enzymes involved in basic cellular functions ranging from cellular respiration to DNA replication, iron is essential for life. Meanwhile, iron can also be harmful, because it readily catalyzes the generation of toxic reactive oxygen species (ROS) from normal products of aerobic respiration in the Fenton reaction. This review summarizes the iron-dependent regulation in bacteria. The primary roles of iron-dependent regulatory proteins Fur (ferric uptake regulator) family and DtxR (diphtheria toxin repressor) family are of special interest in M. tuberculosis. Exploring the regulatory mechanism of iron metabolism in M. tuberculosis, will provide a new way for the treatment of tuberculosis. |
