α-酮戊二酸脱氢酶基因多态性与优秀有氧运动能力的关联研究

摘 要: | 目的:选取α-酮戊二酸脱氢酶系(α-KGDHC/OGDH)两个同工酶OGDH和OGDHL中OGDHL基因的rs1268722位点,比较该位点在我国优秀长跑运动员和普通人中的分布频率,探讨OGDHL基因多态性与优秀有氧运动能力的关联性。方法:使用case-control实验设计,分析rs1268722单核苷酸多态性(single nucleotide polymorphism,SNP)位点在122名优秀长跑运动员和125名普通人对照组中的分布特征。采用基质辅助激光解析/电离-飞行时间(matrix-assisted laser desorption/ionization time-of-flight mass spectrometry,MALDI-TOFMS)质谱技术对SNP位点进行基因解析。结果:1)运动员组和健将组rs1268722位点A等位基因的分布频率显著高于对照组;2)女运动员组A等位基因频率显著高于女对照组;3)马拉松组AA 基因型和A等位基因频率显著高于对照组。结论:OGDHL基因rs1268722位点AA基因型可以作为马拉松专项长跑运动员的有氧运动能力分子标记,但其具体影响机制还需进一步研究。 |

Abstract: | Objective: rs1268722 site in OGDHL gene of two isozymes (OGDH and OGDHL) of α-oxoglutarate dehydrogenase system (α-KGDHC/OGDH) was selected to compare the distribution frequency in excellent long-distance runners and ordinary people, in order to explore the association of polymorphisms in OGDHL gene with the elite aerobic performance. Methods: A total of 122 elite runners and 125 healthy non-athletes were used in this study and the experiment was designed by case-control. The single nucleotide polymorphism (SNP) site of rs1268722 in OGDHL gene was analyzed with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS). Results: 1) The A allele gene frequency of rs1268722 site in athlete group and elite athlete group is significantly higher than control group. 2) The A allele gene frequency in female athlete group is significantly higher than female control group. 3) The frequencies of AA genotype and A allele gene in marathon athletes are significantly higher than controls. Conclusion: The AA genotype of the gene polymorphism loci rs1268722 in the OGDHL gene is associated with the molecular marker in the aerobic performance of elite marathon athletes. The function of the SNP is needed to be further studied. |
