Rho-Rock信号通路阻断剂对脊髓损伤大鼠神经功能修复的作用

摘 要: 目的:探讨Rho-Rock信号通路阻断剂Fasdiual对脊髓损伤(spinal cord injury,SCI)大鼠神经功能修复的作用。方法:选取8周龄SD雄性大鼠36只,采用改良Allen打击法制备SCI大鼠模型后根据实验干预措施随机分组,A组为骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMSCs)移植组、B组为BMSCs移植+Fasdiual组、C组为BMSCs移植+生长因子组。于SCI术后4周采用BBB(Basso-Beattie-Bresnahan)评分法对SCI大鼠后肢运动功能评分后,取脊髓损伤节段制备标本,采用免疫组化染色观察对比两组神经干样细胞特性表达蛋白NF200、NSE及胶质细胞胶质纤维酸性蛋白(glial fibrillary acidic portein,GFAP)的阳性表达。结果:SCI术后4周各组大鼠后肢运动功能评分均有明显改善,B、C组SCI大鼠后肢运动功能评分均优于A组,差异有统计学意义(P<0.05);SCI大鼠脊髓标本免疫组化结果显示NF200、NSE在B、C组表达量均高于A组,差异有统计学意义(P<0.05),GFAP在A组的表达量高于B、C组,差异均有统计学意义(P<0.05)。结论:Rho-Rock抑制剂Fasdiual联合BMSCs定向移植后可促进损伤脊髓的修复,促进SCI大鼠神经功能的恢复。

Title: Effect of Rho-Rock signaling pathway blocker on neurological function recovery in rats with spinal cord injury

Abstract: Objective: To investigate the effect of Rho-Rock signaling pathway blocker (Fasdiual) on neurological function recovery in rats with spinal cord injury (SCI). Methods: Thirty-six SD male rats aged 8 weeks were selected and prepared by modified Allen strike method for SCI rat model, and randomly divided into several groups according to experimental interventions. Group A is bone marrow mesenchymal stem cells (BMSCs) transplantation group, group B is BMSCs transplantation with Fasdiual group, group C is BMSCs transplantation with growth factor group. After 4 weeks of SCI operation, BBB (Basso-Beattie-Bresnahan) scoring method was used to evaluate the recovery of lower extremity motor function of SCI rats. After preparing specimens from SCI segments, immunohistochemical staining was used to observe the positive expression of two neural stem cell characteristic expression proteins (NF200, NSE), and glial fibrillary acidic portein (GFAP) of glial cell. Results: The motor function scores of lower extremities were significantly improved in all groups after 4 weeks of SCI operation. The scores of lower extremity motor function of SCI rats in group B and C were significantly better than those in group A (P<0.05). The immunohistochemistry results of the spinal cord specimens in SCI rats showed that the expression levels of NF200 and NSE in group B and C were higher than those in group A, with statistical significance (P<0.05). The expression of GFAP in group A was higher than that in group B and C. The difference was statistically significant (P<0.05). Conclusion: Rho-Rock blocker Fasdiual combined with BMSCs stereotaxic transplantation can promote the repair of injured spinal cord and the recovery of neurological function in SCI rats.
