将军山全沟硬蜱体内伯氏疏螺旋体的分型

摘 要: 为确定采集于哈尔滨将军山的全沟硬蜱体内是否携带伯氏疏螺旋体(Borrelia burgdorferi)及其分型情况,本研究应用巢式PCR方法对伯氏疏螺旋体进行检测,并进一步应用分型标记基因OspA和groEL进行分型。应用巢式PCR方法在全沟硬蜱体内检测到伯氏疏螺旋体,OspA和groEL的长度分别为287 bp和289 bp,进化分析结果显示,本实验的伯氏疏螺旋体均与GenBank登录的阿弗西尼疏螺旋体(B. afzelii)序列在同一进化分支上。研究结果表明,采集于哈尔滨将军山的全沟硬蜱携带伯氏疏螺旋体,类型为阿弗西尼疏螺旋体(B. afzelii)。

Abstract: In order to determine whether Borrelia burgdorferi is carried in Ixodes persulcatus collected from General hill in Harbin and to identify the type of B. burgdorferi, nested PCR was used to detect B. burgdorferi, and the typing marker genes OspA and groEL were further used for genotyping. B. burgdorferi was detected in I. persulcatus by nested PCR. The lengths of OspA and groEL were 287 bp and 289 bp, respectively. The evolutionary analysis results showed that the sequences of B. burgdorferi in this experiment were all on the same evolutionary branch as the sequences of B. afzelii registered in GenBank. The results indicated that the type of B. burgdorferi carried by I. persulcatus from General hill in Harbin was B. afzelii.
