时间分辨荧光结合微流控芯片技术应用于肌红蛋白的便捷检测

摘 要: 本研究旨在利用微流控芯片解决时间分辨荧光侧流层析法重复性差的问题,将微流控芯片技术与时间分辨荧光免疫测定技术的优势相结合,对肌红蛋白进行便捷检测,极大增强了分析效率,发展免疫分析检测的新方法。该研究利用玻片为载体固定抗体,将玻片预先埋入微流控芯片中,把试剂反应、样品分离、产物检测等过程集中于一块微小尺寸的芯片上,提高了分析过程的自动化程度,且在十分钟内即可完成整个检测过程,分析效率高。另外,微流控芯片能够实现反应时间严格可控,提高了检测可重复性。这是一种基于双抗体免疫夹心法的新型免疫技术,时间分辨荧光微球与肌红蛋白抗体结合后性质稳定,不易发生解离现象,可直接在玻片上检测到强荧光。该方法可作为肌红蛋白定量分析的依据,且检测灵敏度较高,肌红蛋白最低检测限可达3.85 ng/mL,因此适用于肌红蛋白抗原的临床检验。该新型免疫检测方法具有重复性好、操作简单、检测效率高、成本低廉、灵敏度高等特点,因此将时间分辨荧光技术与微流控芯片技术相结合的方式具有巨大的发展前景。

Title: Time-resolved fluorescence combined with microfluidic chip technology for convenient detection of myoglobin

Abstract: This study aims to solve the problem of poor repeatability of time-resolved fluorescence lateral flow chromatography by using a microfluidic chip. By combining the advantages of microfluidic chip technology and time-resolved fluorescence immunoassay technology, the detection of myoglobin is extremely convenient. The analysis efficiency has been greatly enhanced, and new methods of immunoassay detection have been developed. This work uses glass slides as carriers to immobilize antibodies, and pre-embeds the glass slides in a microfluidic chip. It concentrates the reagent reaction, sample separation, product detection and other processes on a micro-sized chip, which improves the automation of the analysis process. The entire detection process can be completed within ten minutes, and the analysis efficiency is high. In addition, the microfluidic chip can realize the reaction time is strictly controlled, which improves the repeatability of detection. This is a new immunological technology based on the double-antibody immunosandwich method. The time-resolved fluorescent microspheres are stable after binding with the myoglobin antibody, and are not easily prone to dissociation. Strong fluorescence can be detected directly on the glass slide. The method can be used as the basis for the quantitative analysis of myoglobin, and the detection sensitivity is high. The minimum detection limit of myoglobin can reach 3.85 ng/mL, so it is suitable for clinical test of myoglobin antigen. The new immunodetection method has the characteristics of good repeatability, simple operation, high detection efficiency, low cost, and high sensitivity. Therefore, the combination of time-resolved fluorescence technology and microfluidic chip technology has great development prospects.
