基于纳米金-多肽-配位金属铱的纳米磷光探针在蛋白酶体活性检测中的应用

摘 要: 本文基于荧光共振能量转移(fluorescence resonance energy transfer,FRET)技术设计构建纳米磷光探针。设计构建一种用于活细胞蛋白酶体实时活性检测的探针。主要设计思路为利用纳米金和铱金属配位化合物之间产生的FRET现象,通过目标蛋白酶的底物多肽连接二者。纳米磷光探针中的上述底物多肽被相应的蛋白酶体特异性识别和切割。FRET供体(铱)配合物与FRET受体(金纳米颗粒)分离,导致FRET现象消失,呈现铱配位配合物的绿色磷光信号。本研究所设计纳米探针针对蛋白酶体(目标蛋白)产生的磷光信号与其酶活性呈正相关,可用于试管和活细胞水平的蛋白酶体活性检测。此外,本研究中所述的酶活检测探针,针对不同的蛋白酶活性检测具有灵活的转变能力,通过多肽序列的替换,可获得目的蛋白酶活性检测磷光探针,大大拓展了其应用。

Title: The application of a nano-luminescent probe based on gold nanoparticle and iridium coordinated peptide in the detection of proteasome activity

Abstract: In this paper, the nano-luminescent probe was designed and constructed based on fluorescence resonance energy transfer (FRET) technology for real-time detection of proteasome activity in living cells. The main design idea is to use the FRET phenomenon between gold nanoparticl and iridium metal coordination compounds, and connect the two through the substrate polypeptide of the target protease. The above-mentioned substrate polypeptides in the nano-luminescent probes are specifically recognized and cleaved by the corresponding proteasome. The separation of the FRET donor (iridium) complex from the FRET receptor (gold nanoparticles) causes the FRET phenomenon to disappear, presenting the green luminescence signal of the iridium coordination complex. The luminescence signal produced by the designed nanoprobe is positively correlated with the activity of proteasome (target protein), which can be used to detect the activity of proteasome at the level of in vitro and living cells. In addition, the enzyme activity detection probe described in this study has a flexible transformation ability for the detection of different protease activities, and the phosphor probe for the detection of targeted protease activity can be obtained through the substitution of polypeptide sequence, significantly expanding its applications.
