转基因菊花遗传稳定性与杂交育种的研究

摘 要: 本研究对获得的转CHS基因地被菊品种‘美矮黄’植株的表现进行观察,对外源基因的遗传稳定性进行了研究,并利用转基因植株对亲本进行了杂交育种。为研究转入基因在转化植株中能否稳定存在并遗传给后代,对转基因株系048进行了无性繁殖和有性繁殖。无性繁殖包括花瓣培养和组培苗多次继代,有性繁殖为对转基因株系048进行植株杂交。对获得的无性繁殖和有性繁殖的后代进行PCR检测,结果表明,转入的外源基因能够在组培苗继代的过程中稳定存在,在花瓣培养和杂交后代中也检测到了外源基因的存在。利用转基因株系048和032的植株在春季进行杂交育种,结果表明,将杂交育种与促成栽培相结合,在温室中进行春季杂交是加速菊花育种的有效方法,可缩短育种周期,获得表现丰富的杂交后代。

Title: Study on genetic stability and cross breeding of the transgenic chrysanthemum (Dendranthema �grandiflorum)

Abstract: In this study, the performances of the CHS transgenic chrysanthemum ‘Meiaihaung’ were observed and the stability of the introduced gene was studied. The transgenic plants were also crossed to get new cultivars. The transgenic line 048 was asexual produced and sexual produced in order to research the stability of the introduced gene in the transgenic plants. In asexual production the petals are tissue cultured. Transgenic line 048 was crossed asexual production. The offspring of both asexual and sexual production were analyzed in the PCR experiments. The results showed that the CHS gene was reserved during the course of propagation. The plants of line 048 and 032 were crossed in the spring in the green house. The result showed that crossing of chrysanthemums in the spring was an effective way to shorten the periods of breeding. The Fl with different characters was obtained.
