使用圆二色性光谱和傅里叶变化红外光谱研究冬小麦麸皮抗冻蛋白的二级结构

摘 要: 本文使用圆二色性光谱和傅里叶变化红外光谱技术对冬小麦麸皮抗冻蛋白的二级结构进行研究。由于冬小麦麸皮抗冻蛋白在酰胺I带和Ⅲ带位置中,各个特征峰相互叠加,因此采用Gausse峰型对圆头峰进行拟合、解析、判断其α-helix,β-sheet,β-turn和random coil的相对含量,并讨论了两种检测方法结果差异的原因,为冬小麦麸皮抗冻蛋白抗冻机理的研究奠定基础。

Title: The secondary structure of winter-wheat-bran antifreeze protein determined by FT-IR and CD spectrum

Abstract: The secondary structure of the winter-wheat-bran antifreeze protein was analyzed by the fourier transform infrared (FT-IR) and circular dichroism (CD) spectrum. The smooth peak of the amide I and amide III region from the FT-IR profile was overlapped by the characteristic peak. So the characteristic peak of the α-helix, β-sheet, β-turn and random coil of winter-wheat-bran antifreeze protein was resolved. The content of the α-helix, β-sheet, β-turn and random coil was the accumulation of the characteristic peak respectively. Moreover, the secondary structure was evaluated by the CD spectrum. Finally, the difference between the FT-IR and CD spectrum results was discussed.
