T细胞识别中复杂现象的新解读

摘 要: T细胞的免疫识别非常特殊,是由T细胞抗原受体(T cell receptor, TCR)和其辅受体(CD4/CD8)共同识别抗原肽-MHC分子(peptide-major histocompatisility complex molecules, pMHC)复合物。这种独特的识别现象可能是一种克隆T细胞识别pMHC存在多种不同识别模式和机制的反映。正常个体内单个初始CD4+T细胞对pMHC的活化识别存在可形成不同生理学效应的规范识别和联体识别两种策略或机制。规范识别以CD4分子作为辅受体与TCR识别同一pMHC,形成识别单体,主要发生在识别同型pMHC时(其中MHC分子为该克隆T细胞阳性选择发育所对应的),与识别耐受有关。联体识别以CD4分子作为连结者识别两(多)个pMHC,主要发生在识别异型pMHC时(其中MHC分子为非阳性选择发育所对应的),形成识别寡聚体,启动增殖分化性应答。T细胞的这种复杂识别可使其能区分“同型(期)/异型(期)”pMHC分子(细胞),有着重大的发育生物学意义。

Title: A novel unscrambling of the complicated phenomena of T cell recognition

Abstract: There are some special characteristics and antinomy phenomena in the complicated T cell antigen recognitions that involve the binding antigen peptide-major histocompatibility complex molecules (pMHC) through the T cell receptor (TCR) with co-receptor (CD4 or CD8). The characteristics and antinomy phenomena, were images of that there were some different recognition patterns and mechanisms in the pMHC recognitions of clone T cells. In a normal individual, a single naive CD4+ T cell recognizing pMHCs may have two different recognizing strategies or patterns, the canonical recognition and the combine recognition, with different physiological consequences. The canonical recognition with CD4 as a co-receptor, usually takes place in recognizing the homo-type pMHCs in which the MHC molecule is the original one during the T cell positive selective development. The canonical recognition forms recognizing monomers and results in recognizing tolerance. The combine recognition with CD4 as a linker, usually takes place in recognizing the hetero-type pMHCs in which the MHC molecule is not the positive selective one. The combine recognition forms a recognizing oligomer and results in multiplication and differentiation responses of the T cells. The complicated recognitions of T cells, which may make them identify the homo-type pMHC or ‘homo-phase’ cell from the hetero-type pMHC or ‘hetero-phase’ cell, have great significance of development biology.
