穹窿切断对大鼠下丘脑室旁核AVP表达的影响

摘 要: 目的:观察切断穹窿大鼠下丘脑室旁核(paraventricular nucleus,PVN)精氨酸加压素(arginine vasopressin,AVP)表达的变化,探讨穹窿切断对丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal,HPA)轴的影响。方法:成年健康雄性Wistar大鼠45只,随机分为穹窿切断0,4,7,10 d组及相应的假手术组和正常组,每组5只。建立大鼠穹窿切断模型,采用免疫组织化学和图像分析技术检测各组下丘脑PVN内AVP的表达变化和分布规律。结果:穹窿切断组于7 d时AVP表达升高,10 d时升高显著。假手术各组和正常组下丘脑PVN仅有少量AVP表达。结论:穹窿切断使下丘脑室旁核AVP表达升高,HPA轴活动增强。

Title: Effects of fornix transection on AVP expression in hypothalamic paraventricular nucleus of rats

Abstract: Objective: To observe the changes of expression of arginine vasopressin (AVP) in the hypothalamic paraventricular nucleus (PVN) of fornix transection rats, and to investigate the effect of fornix transection on HPA axis. Methods: 45 Wistar male rats were randomly divided into fornix transection 0, 4, 7, 10 d groups and corresponding sham-operated groups and the normal group with 5 rats in each group. Set rats fornix transection models. The changes of expression and concentration of AVP in PVN of rats were detected by immunohistochemistry and analysis of images in each group. Results: The expression of AVP in the fornix transection group increased on 7 d and increased obviously on 10 d. There was only a little expression of AVP in PVN in the sham-operated groups and the normal group. Conclusion: The fornix transection can increase the expression of AVP and enhance the activity of HPA axis.
