芍药苷抗大鼠心肌缺血及抗心肌细胞凋亡作用

摘 要: 研究芍药苷对心肌缺血大鼠模型及心肌细胞凋亡的影响,将24只大鼠随机分为对照组、高剂量组、低剂量组及假手术组,建立大鼠心肌缺血模型。实验测定各组心肌缺血梗死范围以及磷酸肌酸激酶(creatine phosphokinase,CPK)和乳酸脱氢酶(lactate dehydrogenase,LDH)指标,并应用脱氧核糖核苷酸末端转移酶介导的缺口末端标记原位细胞凋亡(TUNEL)法测定各组心肌细胞凋亡及凋亡指数,比较各组间差异。实验结果显示:高剂量组较对照组和低剂量组能明显降低心肌缺血大鼠模型的危险指数,抑制心肌中CPK和LDH活性的升高;高剂量组能明显降低心肌细胞的凋亡指数,对心肌细胞凋亡的保护作用明显高于对照组和低剂量组,与假手术组无统计学差异。本研究提示:芍药苷对心肌缺血大鼠模型心肌梗死有明显的保护作用,其作用机制可能与抗心肌细胞凋亡作用有关。

Title: Effects of paeoniflorin on anti-myocardial ischemia in rats and anti-myocardial apoptosis

Abstract: This paper studied the effects of paeoniflorin on anti-myocardial ischemia in rats and anti-myocardial apoptosis. Divided 24 rats into 4 groups randomly with control group, high-dose group, low-dose group, sham-operated group, then established the model of myocardial ischemia rats. Determined the myocardial infarction areas, the values of CPK and LDH of all rats. And detected the myocardial apoptosis with the terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) technique of all rats. Compared the differences between each group, the results showed that the high-dose group could decrease the risk index, the content of CPK and LDH and the apoptosis index of myocardial cells remarkably. The high-dose group had protective effects on myocardial cells. It was higher than the control group and the low-dose group. And the high-dose group had no statistical difference compared with the sham-operated group. The research suggested that paeoniflorin has obviously protective effects on myocardial infarction in the myocardial ischemia rats. The mechanism possibly relates to the effects of anti-myocardial apoptosis.
