半夏泻心汤活性成分对Cajal间质细胞膜电位及线粒体膜电位的影响

| 摘 要： | 目的：观察半夏泻心汤活性成分小檗碱、黄芩苷、甘草次酸和人参皂苷Rg1对小鼠Cajal间质细胞（interstitial cells of Cajal，ICC）膜电位及线粒体膜电位的影响。方法：体外分离培养ICC，用小檗碱、黄芩苷、甘草次酸和人参皂苷Rg1处理ICC，作用24 h后，采用ICC特异的c-kit抗体进行免疫荧光测定，利用激光共聚焦显微扫描技术和特异性荧光探针标记细胞膜电位及线粒体膜电位。结果：半夏泻心汤活性成分中，小檗碱能明显提高ICC膜电位及线粒体膜电位，甘草次酸和人参皂苷Rg1能提高ICC线粒体膜电位，黄芩苷对二者均无明显作用。结论：半夏泻心汤调节胃肠动力的作用机制可能与其活性成分提高ICC膜电位及线粒体膜电位有关。 |

| Abstract： | Objective: Observe the effects of Berberine, Baicalin, Glycyrrhizic acid, and Ginsenoside Rg1 which are the active components of Banxia Xiexin decoction (BD) on the membrane potential (MP) and mitochondrial membrane potential (MMP) of interstitial cells of Cajal (ICC). Methods: The ICC was dissociated and cultured in vitro, then treated with active components for 24 hours, c-kit immunofluorescence antibody was used for its identification under the confocal microscopy, utilized the scanning technique of laser confocal scanning microscope and specificity fluorescent probe to mark the MP and MMP of ICC. Results: Berberine can enhance the MP and MMP of ICC. Glycyrrhizic acid, Ginsenoside Rg1 can increase the MMP of ICC. Baicalin has no significant effect on them. Conclusion: The mechanism of BD regulates the gastrointestinal motility may be through these active components enhancing the MP and MMP of ICC. |
