小平台水环境间断睡眠剥夺法复制大鼠失眠模型的评价

摘 要: 目的:小平台水环境间断睡眠剥夺法复制大鼠失眠模型的评价。方法:运用小平台水环境间断睡眠剥夺法建立模拟失眠大鼠模型,观察其对大鼠的自主活动、睡眠脑电反映的睡眠时相和睡眠相关物质5-HT、NE的影响。结果:模型组大鼠与对照组比较,其自主活动次数明显增多(P<0�01);睡眠时相觉醒(wake)期时间明显延长,而总睡眠时间(total sleep time, TST)明显缩短(P<0�05);慢波睡眠(slow wave sleep, SWS)时间明显减少,其中慢波睡眠Ⅱ期(slow wave sleep Ⅱ,SWS2)减少最为显著(P<0�01),而慢波睡眠Ⅰ期(SWS1)无明显变化;快眼动睡眠(rapid eye movement sleep, REMS)有所增加,但无统计学意义(P>0�05)。结论:小平台水环境间断睡眠剥夺法,可增加觉醒、减少睡眠;增加大鼠的兴奋性;影响睡眠相关物质(5-HT,5-HIAA,NE)的昼夜节律,使其分泌失衡,使大鼠睡眠-觉醒紊乱,较好地模拟了失眠的病理状态。且此造模方法恢复期间可多次给药,满足中药作用时间慢、治疗周期长、镇静催眠作用弱、注重“调理”而非强调“重镇”的特点,能较好地适用于中药制剂治疗失眠的动物实验研究。

Abstract: Objective: The evaluation of the insomnia rat which was reproduced by placing rats on a small platform suspended water by interruptable sleep deprivation. Methods: The insomnia rat was reproduced by using a sleep-disturbed model developed by placing rats on a small platform suspended water by interruptable sleep deprivation. The responses of the insomnia rat to time choosing-homeopathic remedy were recorded in following aspects: the general condition, the autonomic activities, study and memory ability, sleep phase of electroencephalogram, sleep related materials such as 5-HT, NE. Results: The treatment group in comparison with the controls show that the independent activity increased considerably (P<0.01). Time of the sleep phase of wake was extended considerably and the sleep phase of TST was shortened considerably (P<0.05). Time of the sleep phase of SWS was shortened considerably, most strikingly shortened was the SWS2 (P<0.01) and the SWS1 without significantly changing; the sleep phase of REMS was increased but it had no statistically significance (P>0.05). Conclusion: The insomnia rat was reproduced by using a sleep-comdisturbed model developed by placing rats on a small platform suspended water by interruptable sleep deprivation. It can promote the disenchantment and reduce the sleep, increase the excitability of the rats, and make the secretion of sleep related materials such as 5-HT, 5-HIAA, NE unbalanced. The method which reproduced the model can respond the pathology of the insomnia better and the drug can be used in the convalescence. It can respond the characteristics of the Chinese traditional medicine such as cure slowly, long therapeutic procedure and calm or enter sleep unconspicuously. The Chinese traditional medicine pays more attention to the regulation and control but not the calm. This method can be used in the investigation of the Chinese traditional medicine which cures the insomnia animals.
